a b s t r a c tPlant serine proteases have been widely used in food science and technology as well as in medicine. In this sense, several plant serine proteases have been proposed as potential anti-coagulants and antiplatelet agents. Previously, we have reported the purification and identification of a plant serine protease from Solanum tuberosum leaves. This potato enzyme, named as StSBTc-3, has a molecular weight of 72 kDa and it was characterized as a subtilisin like protease. In this work we determine and characterize the biochemical and medicinal properties of StSBTc-3. Results obtained show that, like the reported to other plant serine proteases, StSBTc-3 is able to degrade all chains of human fibrinogen and to produces fibrin clot lysis in a dose dependent manner. The enzyme efficiently hydrolyzes b subunit followed by partially hydrolyzed a and g subunits of human fibrinogen. Assays performed to determine StSBTc-3 substrate specificity using oxidized insulin b-chain as substrate, show seven cleavage sites: Asn3-Gln4; Cys7-Gly8; Glu13-Ala14; Leu15-Tyr16; Tyr16-Leu17; Arg22-Gly23 and Phe25-Tyr26, all of them were previously reported for other serine proteases with fibrinogenolytic activity. The maximum StSBTc-3 fibrinogenolytic activity was determined at pH 8.0 and at 37 C. Additionally, we demonstrate that StSBTc-3 is able to inhibit platelet aggregation and is unable to exert cytotoxic activity on human erythrocytes in vitro at all concentrations assayed. These results suggest that StSBTc-3 could be evaluated as a new agent to be used in the treatment of thromboembolic disorders such as strokes, pulmonary embolism and deep vein thrombosis.

In the present investigation antibacterial activity of latexes from ten Indian plant species Spondias dulcis (Amra), Diospyros melanoxylon (Tendu), Terminalia bellirica (Wahera), Ficus glomerata (Gular), Phyllanthus emblica (Awla), Thevetia neriifolia (Kaner), Carica papaya (Papita), Calotropis procera (Ak), Ficus benghalensis (Bargad), Artocarpus heterophyllus (Kathal) collected from Gorakhpur, North India were determined in various in vitro systems. MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration) values were determined in growth inhibitory bioassays by using different increasing concentrations of various latex extracts. Latex samples were diluted by using serial micro dilution method up to 10 −10 method with Luria broth culture medium. These values were obtained significantly 2 -3 times lower than that of broad spectrum antibiotic drugs. Besides this, inhibition zone diameters were measured in agar disc diffusion assay. A known volume i.e. 0.1 -20 μg/μl of each latex were coated on separate sterile filter paper discs (Whatman No. 1) measuring 6 mm in size. Latex fractions registered significantly higher growth inhibition than that of broad spectrum antimicrobial drugs. Present study indicates the potential use of shows that both latex and its components and latex as are valuable source of medicinal products/active principles that can be used for treatment of life threatening infectious diseases. Because of higher inhibitory and cidal potential obtained in latexes than the synthetic drugs these that could lead to become efficient phytomedicines mainly to have and develop as complete drug formulations against to control infectious microbes.

Objective: The current study assesses the fibrinogen and fibrin clot hydrolyzing activities of aqueous seed extract of Jackfruit (AqSEJ). Methods: The protein banding pattern of AqSEJ (100 μg) was analyzed on SDS-PAGE. The proteolytic activity of AqSEJ was confirmed by spectrophotometer and zymography experiments. Fibrinogen, fibrin and plasma protein hydrolyzing activities of AqSEJ were analyzed on SDS-PAGE under reduced conditions. Plasminogen activation and indirect hemolytic activities was analyzed using spectrophotometer. The non-toxic property of AqSEJ was tested by edema, hemorrhage in experimental mice. Results: AqSEJ exhibited proteolytic activity and the specific activity was found to be 1.04 units/mg/min. Furthermore, AqSEJ non-specifically hydrolyzed Aα, followed by Bβ and γ chains of human fibrinogen and specifically hydrolyzed α polymer and α chain of partially cross linked human fibrin clot without affecting β chain and γ-γ dimer even up to the tested dose of 30 μg for the incubation period of 8 hours. Importantly, AqSEJ did not hydrolyze other plasma proteins and devoid of plasminogen activation property. The proteolytic activity of AqSEJ was completely neutralized by PMSF and IAA, while EDTA, EGTA, 1,10-Phenanthroline did not, suggesting the presence of serine and cysteine family proteases. Moreover, AqSEJ did not cause edema and hemorrhage in experimental mice up to the tested dose of 200 μg and non-toxic to RBC cells. Conclusion: AqSEJ hydrolyzes fibrinogen and fibrin clot and non-toxic in nature. Hence, this work showcases the potential applications of Jack fruit seed proteases in the treatment of thrombotic disorders.
