Processing and secretion of insulin-related peptides in an insulinoma cell line

Abstract: Certain classes of prohormones and other neuroendocrine or endocrine‐derived secretory proteins are post‐translationally modified in the secretory storage granules. If such molecules were to be biosynthesized to acceptable quantity and yield using endocrine‐derived cell lines, it would be important to understand the relationship between the secretory dynamics and the conversion and release of the immature and mature forms of the molecule. We studied aspects of such a relationship using the endocrine‐derived ce… Show more

“…The manufacturer's reported conversion between standard insulin activity units and equivalent grams of mature human insulin was approximately 27.6 μU/ng insulin. High‐performance liquid chromatography (HPLC) analysis of secreted and intracellular peptides from βTC‐3 cells confirmed that significant amounts of mature insulin and proinsulin are present in the samples (Grampp et al, 1996b). Accordingly, all RIA data reported include contributions from both species and are reported as units of total IRPs.…”
