Procedure for detecting tobamovirus in tomato and pepper seeds decreases the cost analysis

Abstract: The transmission of tobamovirus by tomato and pepper seeds is an important mean of virus introduction in crops. Therefore, detecting its presence in the seed becomes essential for the preventive control of virus diseases. In this study, a method was proposed for the detection of Tobacco mosaic virus (TMV) and Tomato mosaic virus (ToMV) in tomatoes (Solanum lycopersicum) and Pepper mild mottle virus (PMMoV) in pepper (Capsicum annum) seeds. Seed lots with different levels of incidence were analyzed by biologica… Show more

“…Biological, serological, and molecular assays have generally been used for identification of plant viruses. Although ELISA is preferred assay for routine virus detection, RT-PCR has increasingly been used for detection and identification of viruses due to higher level of sensitivity (Almeida et al, 2018). Among the molecular methods, the most widely used is the RT-PCR (Liu et al, 2014).…”

“…The ELISA tests used in the diagnosis work for many plant viruses have widely been used by researchers (Geyik, 2017;Kapoor et al, 2018). The fact that it is fast, sensitive, economical and reliable makes this method a preference (Vinayarani et al, 2011;Almeida et al, 2018).…”

“…Sap transmission was conducted by crushing 0.1 g of diseased leaf with 1 mL buffer phosphate 0.005 M pH 7 and 10 µL mercaptoethanol. Sap was inoculated into the Chenopodium amaranticolor and Nicotiana benthamiana by mechanical inoculation (Almeida et al, 2018). The virus morphology was visualized using transmission electron microscope (JEOL JEM 1400, Japan) by a quick dipping method with negative stain using ammonium molybdat 1% pH 7 (Gebre-Selassie & Marchoux, 1991;Latifah et al, 2008).…”
