Probing Reversible Guest Binding with Hyperpolarized 129Xe-NMR: Characteristics and Applications for Cucurbit[n]urils

Abstract: Cucurbit[n]urils (CB[n]s) are a family of macrocyclic host molecules that find various applications in drug delivery, molecular switching, and dye displacement assays. The CB[n]s with n = 5–7 have also been studied with 129Xe-NMR. They bind the noble gas with a large range of exchange rates. Starting with insights from conventional direct detection of bound Xe, this review summarizes recent achievements with chemical exchange saturation transfer (CEST) detection of efficiently exchanging Xe in various CB[n]-ba… Show more

“…Notably, the IDA format is also applicable to other signal transduction methods besides absorbance and fluorescence spectroscopy. For instance, 129 Xe can be used as an NMR indicator to follow in vitro and in vivo binding of spectroscopically silent analytes to CB6 and CB7, as has been demonstrated by the Dmochowski group, the Albert group, and by Schröder, Hennig, and coworkers. …”

“…However, their commercial unavailability, complex synthesis involving multiple steps, and low yields pose some challenges. While cryptophane-A is difficult to synthesize, the biocompatible and readily available cucurbit [6]uril [104,167,168] has been recognized as a promising alternative. In the next subsection, we delve into cucurbit [6]uril's biosensing properties.…”

“…Naturally, this is reflected in its NMR signal, which, analogously to solid materials, can be used to obtain in-depth structural and morphological information along with knowledge about phase separation, dynamics, and presence and size of different domains. Owing to these important features, 129 Xe NMR has been used to study a variety of pure liquids, mixtures, solutions, and host-guest complexes in the liquid phase [51][52][53][54][55][56][57].…”
