Proapoptotic activity of indoleamine 2,3-dioxygenase expressed in renal tubular epithelial cells

Abstract: Exposure of renal tubular epithelial cells (TEC) to IFN-gamma/TNF-alpha leads to Fas/FasL-mediated self-injury, which contributes to allograft rejection. Indoleamine 2,3-dioxygenase (IDO) converts tryptophan to N-formyl-kynurenine and contributes to immune privilege in tissues by increasing Fas-mediated T cell apoptosis. However, renal expression of IDO and its role in promoting Fas-mediated TEC death have not been examined. IDO expression was analyzed by RT-PCR and Western blot. Apoptosis was measured by fluo… Show more

“…Renal IRI causes severe tissue injury and various forms of cell death [4,5,6,7,8] including apoptosis, necrosis, autophagy, and other nonclassical forms of cell death [42]. During necrotic cell death, cells invariably lose membrane integrity and eventually lyse, resulting in the release of intracellular contents and various DAMP such as HMGB1.…”

“…The TEC line NG1.1 was developed from proximal TEC of C57BL/6J mice by SV40 transformation as previously described [4]. NK cells were purified from C57BL/6J mouse spleens using anti-CD49a MACS bead selection (Miltenyi Biotec) and were grown in the presence of IL-2 (1,000 IU/ml) in RPMI-1640.…”

“…Given that isc DCs appear to migrate more rapidly to recipients LNs, it follows that a more robust recipient T cell response would occur and that graft survival would be accordingly more compromised. Our observation that isc DCs undergo significantly more apoptosis than ctrl DCs provide a further mechanism by which isc grafts display greater immunogenicity, as apoptosis and necrosis of donor cells have been implicated in the established increased immunogenicity of isc allografts (77)(78)(79)(80)(81)(82). DCs have also been specifically implicated in acquiring Ag from apoptotic cells to stimulate class Irestricted cytotoxic T cells (83,84).…”

“…These findings are of particular interest given animal and cellular studies that implicate tryptophan metabolism through the kynurenine pathway as a mediator of renal injury (4,5), and demonstrate functional roles to kynurenine and kynurenic acid in vascular tone and inflammation (6,7). Other risk markers identified in the FHS included citrulline (OR per 11.48 SD, 1.38; P,0.001; 95% CI, 1.19 to 1.83; [3]) and choline (OR per 11 SD, 1.46; P,0.001; 95% CI, 1.17 to 1.82; [3]).…”
