Pro-Inflammatory Signalling PRRopels Cisplatin-Induced Toxicity

Abstract: Cisplatin is a platinum-based chemotherapeutic that has long since been effective against a variety of solid-cancers, substantially improving the five-year survival rates for cancer patients. Its use has also historically been limited by its adverse drug reactions, or cisplatin-induced toxicities (CITs). Of these reactions, cisplatin-induced nephrotoxicity (CIN), cisplatin-induced peripheral neuropathy (CIPN), and cisplatin-induced ototoxicity (CIO) are the three most common of several CITs recognised thus far… Show more

“…However, due to the resistance of patients to CP, conventional therapy has a disappointing effect [6]. Besides, CP also has systemic toxicity, including central nervous system damage and nephrotoxicity [7]. All of these factors limit its application in the treatment of melanoma.…”

“…Additionally, cisplatin induced neurotoxicity with a severe inflammatory and proinflammatory mediator’s induction, such as TNF α and NF-κB [ 201 , 202 ]. TNF-α is an essential mediator of chronic inflammation and a significant contributor to peripheral nerve injury and neuropathic pain [ 203 , 204 ].…”

“…Inflammation is also reported to be involved in developing the side effects of chemotherapy [ 58 ]. Chemotherapy-induced toxicity of cisplatin has been reported to be enhanced by inflammation [ 59 , 60 ]. Therefore, the presence of inflammation before chemotherapy may influence chemotherapy discontinuation due to the side effects of the anticancer drugs.…”
