Prion protein expression in different species: Analysis with a panel of new mAbs

Abstract: A BSTR ACTBy immunizing prion knockout mice (Prnp؊/؊) with recombinant murine prion protein (PrP c ), we obtained a panel of mAbs specific for murine PrP c . These mAbs can be applied to immunoblotting, cell surface immunof luorescent staining, and immunohistochemistry at light and electron microscopy. These mAbs recognize both the normal (PrP c ) and protease-resistant (PrP res ) isoforms of PrP. Some mAbs are species restricted, while others react with PrP from a broad range of mammals including mice, humans… Show more

“…Anti-PrP monoclonal antibody 3F4 (specific to PrP residues 109 -112) was from Richard Kascsak (New York State Institute for Basic Research in Developmental Disabilities). Anti-PrP monoclonal antibody 8H4, which binds to a site between residues 145-180 of PrP, was raised in our facility (20). All other chemicals, including antibodies to the glial fibrillary acidic protein (anti-GFAP) and neurofilament-specific (NF68) antibodies were purchased from Sigma.…”

“…The generation of recombinant PrP C proteins and anti-PrP monoclonal antibodies (MAbs) was described in detail previously (20,49,50). All MAbs were affinity purified.…”

“…Mouse monoclonal antibody 8H4, epitope (residues 145-220) binding is independent of the N-linked glycosylation, because it reacts with both recombinant PrP and all native glycoforms (29). 7A12 (epitope 90 -140) is a mouse monoclonal anti-PrP antibody (30).…”
