The cancer killing efficacy of standard chemotherapeutic agents such as cisplatin (CDDP) is limited by their side effects to normal tissues. Therefore, research efforts optimizing the safety and efficacy of those agents are clinically relevant. We did screen for agents that specifically protect normal human mesothelial cells against CDDP without reducing the cancer cell killing efficacy. Lovastatin was identified from the screen. Lovastatin at a pharmacologically relevant concentration strongly arrested the proliferation of normal cells, whereas cancer cells were less affected. CDDP-induced DNA damage response was not activated and normal cells showed enhanced tolerance to CDDP when normal cells were treated with the combination of CDDP and lovastatin. We demonstrate that interfering with protein geranylgeranylation is involved in the lovastatin-mediated CDDP protective effect in normal cells. In contrast to normal cells, in cancer cells lovastatin did not change the CDDP-induced response, and cancer cells were not protected by lovastatin. Furthermore, lovastatin at the pharmacological relevant concentration per se induced DNA damage, oxidative stress and autophagy in cancer cells but not in normal mesothelial cells. Therefore, our data suggest that lovastatin has a potential to improve the therapeutic index of cisplatin-based therapy.

Nanoscale materials have been explored extensively as agents for therapeutic and diagnostic (i.e. theranostic) applications. Research efforts have shifted from exploring new materials in vitro to designing materials that function in more relevant animal disease models, thereby increasing potential for clinical translation. Current interests include non-invasive imaging of diseases, biomarkers and targeted delivery of therapeutic drugs. Here, we discuss some general design considerations of advanced theranostic materials and challenges of their use, from both diagnostic and therapeutic perspectives. Common classes of nanoscale biomaterials, including magnetic nanoparticles, quantum dots, upconversion nanoparticles, mesoporous silica nanoparticles, carbon-based nanoparticles and organic dye-based nanoparticles, have demonstrated potential for both diagnosis and therapy. Variations such as size control and surface modifications can modulate biocompatibility and interactions with target tissues. The needs for improved disease detection and enhanced chemotherapeutic treatments, together with realistic considerations for clinically translatable nanomaterials will be key driving factors for theranostic agent research in the near future.

RESUMENObjetivo. Evaluar los valores medios del hemograma en perros sanos durante la quimioterapia con cisplatina. Materiales y métodos. Se evaluaron los parámetros del hemograma de ocho caninos sanos sometidos a terapia con cisplatina, distribuidos en dos grupos. A los perros del grupo uno se les administró cisplatina (70 mg/m², i.v.) y para prevenir la nefrotoxicidad recibieron suero fisiológico al 0.9% (25mL/kg/h, i.v.). Los animales del grupo dos no recibieron cisplatina. Resultados. El hemograma de los perros del grupo uno no mostró diferencia significativa (p>0.05) con respecto al de los animales del grupo dos. Conclusiones. Los datos obtenidos indicaron que el protocolo con cisplastina no alteró significativamente los valores medios del hemograma y el número de sesiones de quimioterapia aumenta el riesgo de disminución de los valores del hemograma.Palabras clave: Cisplatina, quimioterapia, hemograma, hemoglobina, leucocitos, perros. ABSTRACTObjective. To evaluate the average values of the hemogram test in healthy dogs during chemotherapy with cisplatin. Materials and methods. The hemogram parameters of eight healthy canines, which underwent therapy with cisplatin, were evaluated. These canines were divided into two groups. The dogs in the first group were given cisplatin (70mg/m², i.v.) and to prevent kidney poisoning they were given physiological serum at 0.9% (25mL/ kg/h, i.v.). The dogs in group two were not given cisplatin. Results. The hemogram of the dogs in the first group did not show any significant difference (p>0.05) as compared to the dogs in group 2.Conclusions. The data obtained indicate the protocol used with cisplatine does not significantly alter the average values of the hemogram test and the number of chemotherapy sessions increases the risk of diminishing the values of the hemogram test.
