Prime/boost immunotherapy of HPV16-induced tumors with E7 protein delivered by Bordetella adenylate cyclase and modified vaccinia virus Ankara

Abstract: The Bordetella adenylate cyclase toxoid (CyaA) targets cells expressing the alphaMbeta2 integrin receptor CD11b/CD18 (CR3 or Mac-1) and can penetrate into cytosol of professional antigen-presenting cells, such as dendritic cells. This allows us to use CyaA for delivery of passenger antigens into the cytosolic pathway of processing and MHC class I-restricted presentation, which can promote induction of antigen-specific CD8+ cytotoxic T-lymphocyte immune responses. We show here that vaccination with a geneticall… Show more

“…The antigen delivery systems most commonly used to induce protective immunity against transformed cells are based on administration of plasmids by gene gun or inoculation of viral vectors containing the gene of interest. Heterologous prime-boost schemes, where DNA and viruses are inoculated in succession, often induce the best immunity (19,28).…”

“…Immunization of mice with genetically detoxified CyaA bearing appropriate T-cell epitopes provides full prophylactic protection against lethal lymphocytic choriomeningitis virus challenge (33), efficient therapeutic antitumor immunity (11) and complete therapeutic efficacy against tumor cells expressing an immunogenic oncoprotein of human papillomavirus 16 in a mouse experimental model mimicking human papillomavirus-induced cervical cancer (21,30).…”
