Primary Renal Cell Carcinoma in a Transplanted Kidney: Genetic Evidence of Recipient Origin

Abstract: We report the first case of a grafted-kidney RCC whose recipient's cell origin has been proved by microsatellite analysis. An origin from the recipient's kidney or bone marrow stem cells is proposed as the more plausible hypothesis.

“…Immunohistochemistry and hematoxilin-eosin staining on formalin-fixed paraffin-embedded tissue sections were performed as described (23). For immunohistochemistry, we have optimized the signal background for anti-PADI2 (Proteintech UK; reference number 12110-1-AP) using positive and negative control samples.…”

“…Lysis and Western blot analyses were performed as previously described (9,42). Immunohistochemistry and immunofluorescence on paraffin-embedded sections were performed as described previously (5,24). For the separation of nucleosolic and chromatin fractions, we have used a modified version of the Dignam protocol.…”

“…Once RCC in the transplanted kidney is diagnosed, it is crucial to determine the genetic origin of the tumor by means of DNA analysis. Thus the potential transmission of tumor cells to other recipients from the same donor can be assessed (Boix, et al, 2009). There is no consensus on treatment of RCC in the transplanted kidney.…”
