Abstract: Although primary nodular amyloidosis is a rare entity it seems to be the most frequent presentation of penile-limited cutaneous disease, with a low incidence of systemic involvement.

“…2 In our case, however, immunohistochemical findings were positive for 34ßE12, a monoclonal antibody against certain high-molecularweight keratins (cytokeratins 1, 5, 10, 14) that reacts strongly with epidermal keratinocytes 4 and also with the amyloid in macular amyloidosis and lichen amyloidosus. 5 In addition, there was no specific immunostaining for κ or λ light chains.…”

“…Our current notion of nodular amyloidosis as immunoglobulin derived is, in fact, based on a small number of individual case reports or case series. 2 The accumulating evidence of a keratinocyte origin for at least some cases of nodular amyloidosis suggests that its causes may be more heterogeneous than previously believed. Management of this condition can be difficult.…”

“…2 -4,6-8,11,13 There are three additional cases of penile amyloidosis involving not the glans but the shaft 9,13 (one of these is Case 2 in the report by Merika et al). 3 Lesions varied from papules 3 to plaques 1 -5,7,12 and nodules. 3,6,8 In all cases, amyloid was investigated with Congo red, and in some cases, immunohistochemical studies were also used.…”

“…Approximately 12 cases have been reported in the literature, with the common presentation of asymptomatic, nodular, tan/ yellow, and rubbery deposits on the skin of the penis. 69 Of the 3 cases in a report characterized by IHC, 2 were serum amyloid-associated protein, and the other was an AL-k amyloid; the remaining cases were unable to be fully characterized by IHC or mass spectrometry. None were found to be definitively associated with systemic disease.…”
