Primary analysis and screening of microRNAs in gastric cancer side population cells

Abstract: There are key miRNAs expressed within the SP cells of the gastric cancer cell line MKN-45, and include hsa-miR-3175, hsa-miR-203, hsa-miR-130a, hsa-miR-324-5p, hsa-miR-34a, and hsa-miR-25-star.

“…MiR‐324‐5p was increased in the side population (SP) cells of the gastric cancer cell line MKN‐45 and has been suggested to play an important role in maintaining and regulating the characteristics of SP cells . However, its specific function and mechanism in gastric cancer are not completely understood.…”

“…MiR-324-5p was increased in the side population (SP) cells of the gastric cancer cell line MKN-45 and has been suggested to play an important role in maintaining and regulating the characteristics of SP cells. [10] However, its specific function and mechanism in gastric cancer are not completely understood. MiRNAs existed in RNA-induced silencing complex (RISC) could inhibit the translation and induce degradation of the target mRNAs through interacting with mRNA 3 0 -untranslated regions (3 0 -UTR).…”

“…Previous study reported that MiR‐3175 could promote cell proliferation, invasion and inhibit apoptosis via regulating its target gene HOXB1 in glioma . However, the role of MiR‐3175 is still controversial in carcinogenesis and pathogenesis; for instance, MiR‐3175 was also found to be upregulated in prostate cancer as an oncogenic miRNA, but downregulated in gastric cancer as a tumor suppressor. To the best of our knowledge, in this study we demonstrated for the first time that MiR‐3175 could promote proliferation, migration, invasion, and EMT of cultured human conjunctival epithelial cells by inhibition of Smad7 directly.…”

“…Using a computer-assisted bioinformatics analysis, we predicted that miR-3175 directly targets and suppresses HOXB1 expression. A recent study found that miR-3175 acts as a tumor suppressor in gastric cancer [ 33 ], and microarray profiling demonstrated its association with chronic congestive heart failure [ 34 ]. However, little is known about the effects of miR-3175 on other kinds of human cancer, including glioma.…”
