Preventive efficacy of oral moxidectin at various doses and dosage regimens against macrocyclic lactone-resistant heartworm (Dirofilaria immitis) strains in dogs

Abstract: Background Moxidectin has previously shown limited efficacy (≤ 44.4%) against confirmed macrocyclic lactone (ML)-resistant Dirofilaria immitis strains at 3 µg/kg after single and multiple oral dosages. Three studies were conducted to evaluate higher oral moxidectin doses for efficacy against confirmed ML-resistant D. immitis strains. Methods Dogs were inoculated with 50 D. immitis L3 and randomly allocated to treatments. Study 1: 6 g… Show more

“…Moxidectin at an oral dose of 3 µg/kg has been used for decades for the prevention of heartworm disease caused by D. immitis in dogs. This dose showed 100% efficacy against recent, susceptible field isolates, while at the 24 µg/ kg dosage moxidectin provided improved efficacy against field isolates from the USA that were resistant to macrocyclic lactones [21]. In addition to the prophylactic efficacy against cardiovascular nematodes, the combination of moxidectin, sarolaner and pyrantel in Simparica Trio ™ also provides effective treatment of flea, tick and gastrointestinal nematode infections [22][23][24][25].…”

“…The macrocyclic lactone (ML) class of anthelmintics are the only drug class widely available as heartworm preventatives and are highly effective in the prevention of canine heartworm disease [4]. These drugs work, likely in concert with the dog's immune system, by killing the immature third (L 3 ) and fourth (L 4 ) larval stages of D. immitis thus preventing development to adult worms [5].…”

Lack of efficacy (LOE) of ML heartworm preventatives can result from failure to regularly administer the product according to the approved label directions [4]; however, lack of efficacy due to D. immitis strains resistant to MLs is a growing concern [4].…”

“…The isoxazoline class of ectoparasiticides such as sarolaner, afoxolaner, fluralaner and lotilaner are highly efficacious and have broader spectrum activity against ticks, fleas and/or mites [1][2][3][4][5][6][7][8][9]. The combination of the isoxazolines with one or more additional active ingredients has provided activity against other parasites, such as heartworm and gastrointestinal nematodes [10][11][12][13][14]. Heartworm disease caused by Dirofilaria immitis is endemic in Australia [15][16][17][18].…”
