Tetrahydroxy stilbene glucoside (TSG), an active component from medicinal herb Polygonum multiflorum Thunb, could block the activity of the tissue reninangiotensin system (RAS), which plays a critical role in development of diabetic osteoporosis. This study aimed to determine if TSG therapy could alleviate bone deteriorations in diabetic mouse model induced by streptozotocin. The diabetic mice showed the loss of trabecular bone mass and the changes of trabecular bone microarchitectural parameters as well as the increase in amount of tartrate-resistant acid phosphatase (TRAP)-positive osteoclasts at the distal metaphysis of femur when compared with those of nondiabetic mice. Treatment with TSG significantly elevated calcium content in serum and bone and improved biological parameters of trabecular bone, accompanied by increasing messenger RNA (mRNA) expression of RUNX-2, COL-I, and OCN and protein expression of β-catenin as well as down-regulating protein expression of RAS components including renin and AT1R. In addition, TSG repressed diabetes-induced decrease in ratio of OPG/RANKL expression and increase in sclerostin expression in bone. The similar effects of TSG on osteoblasts-specific genes were found in MC3T3-E1 cells. Taken together, the present study demonstrated the osteopreserve effects of TSG in diabetic mice, and the underlying mechanism might be attributed to its regulation on osteogenesis and osteoclastogenesis.

An increase in anaerobic (oxygen-deprived) pathogenesis significantly increases the generation of reactive oxygen species (ROS) inflicting damage on cell membranes and intracellular constituents. Generation of ROS and concomitant inflammatory response is the two hallmarks of cellular damage caused by cellular injury or invasion by pathogens. Oxygen deprivation, as opposed to oxygen deficiency, is a major contributor to oxidative stress and damage, cytokine production, and inflammation. When our cells are unable to efficiently and effectively utilize the oxygen to facilitate aerobic glycolysis and other cellular metabolic events, the oxygen instead oxidizes cell membranes, lipids, neurons, cross-links proteins, damages DNA, and initiates inflammation among other consequences. These anaerobic events are hallmarks of chronic degenerative diseases (CDD). Excessive demands to curtail oxidative damage can overburden endogenous antioxidative capabilities. A key treatment strategy to tackle the adverse effects of inflammation involves the augmentation of the structural integrity and functional competence of cellular materials, reducing the impact and consequences of tissue insult; the generation of ROS; and the cascade of subsequent pathological disorders. Moreover, restoration of cellular aerobic metabolic events, such as aerobic glycolysis and oxidative respiration, is an equally important collateral goal. A healthy diet and supplementation, providing an abundance of exogenous sources of antioxidants and a host of phytochemical dietary components, becomes even more important to restore aerobic metabolism; augment and assist in improving cellular structural integrity, and thereby reducing oxidative stress, damage, and inflammatory sequela. VMP35 MNC, a research-affirmed Prodosomed nutraceutical technology-based phytonutrient formulation, enriched in structurally diverse bioflavonoids, polyphenols, and phenolic saccharides, etc., have been shown to boost cellular structural integrity and physiological functions, and restore aerobic metabolic competence including for athletic performance as well as for general well-being. Clinical and experimental data have shown that prolonged exposure to GCs leads to bone loss and increases fracture risk. Special attention has been given to existing emerging drugs that can prevent and treat glucocorticoid-induced osteoporosis GIOP. However, there is no consensus about the most relevant animal model treatments on GIOP. In this systematic review, we aimed to examine animal models of GIOP centering on study design, drug dose, timing and size of the experimental groups, allocation concealment, and outcome measures. The present review was written according to the PRISMA 2020 statement. Literature searches were performed in the PubMed electronic database via Mesh with the publication date set between April, 2011, and February 2021. A total of 284 full-text articles were screened and 53 were analyzed. The most common animal species used to model GIOP were rats (66%) and mice (32%). In mice studies, males (58%) were preferred and genetically modified animals accounted for 28%. Our work calls for a standardization of the establishment of the GIOP animal model with better precision for model selection. A described reporting design, conduction, and selection of outcome measures are recommended.
