Preventive Effect of Dapsone on Renal Scarring following Mannose-Sensitive Piliated Bacterial Infection

Abstract: Renal scarring has been thought to occur in the later stages of chronic pyelonephritis. We previously reported that mannose-sensitive (MS) piliated bacteria promoted renal scarring, which was prevented by antioxidants. The preventive effect of diaphenylsulfone (dapsone), which has a scavenging activity on active oxygen species, on renal scarring was examined. Female Sprague-Dawley rats were inoculated with clinical isolates of Serratia marcescens which had both MS and mannose-resistant pili or with recombinant… Show more

“…Other studies with oxytocin [28], melatonin [24,27], pentoxifylline [36], vitamin A [37], and montelukast [30] (a selective leukotriene CyslT1 receptor antagonist) showed the same result. Anti-inflammatory drugs such as glucocorticoids [23,31,32], ulinastatin [38], ibuprofen [25], and dapsone [39] significantly inhibited renal scarring in animal models. However, few researches noted the failure of dexamethasone [26] and hydrocortisone [40] to prevent renal damage following acute pyelonephritis.…”

“…From the report by Yagmurlu et al, it is showed that anticytokine activity of pentoxifylline could be the other mechanism for the prevention of renal scarring due to pyelonephritis, though this study did not include cytokine measurements [9]. The preventive effect of dapsone which has a scavenging activity on active oxygen species on renal scarring was found to effectively prevent renal scarring by Mochida et al [16]. Caffeic acid phenethyl ester (an active component of propolis from honeybee hives, which has antioxidant, anti-inflammatory, and antibacterial properties) administration reduced significantly decreased E. coli -induced lipid peroxidation as showed by Celik et al [20].…”

“…Therefore, ongoing research projects are underway to find an agent that can prevent renal scarring and subsequent complications. Inhibition of acute inflammation in experimental studies by steroids [3, 5], anti-inflammatory agents [6, 7], melatonin [8], pentoxifylline [9], vitamin A [10], vitamins A and E [11], vitamins C and E [12], vitamin E [13], mesenchymal stem cell [14], methylene blue [15], dapsone [16], ulinastatin [17], and montelukast [18] have been reported to reduce kidney damage after infection.…”

“…Dapson, as a free oxygen radical scavenger, prevents renal scar formation with a dose of 20 mg/kg in the experimental pyelonephritis model [36] . Ulinastatin is an elastase inhibitor presented in PMN leukocytes.…”
