Prevention of Activated Neutrophil Adhesion to Endothelium by Soluble Adhesion Protein GMP140

Abstract: Neutrophils and monocytes, but not lymphocytes, adhered strongly to plastic surfaces coated with GMP140, a protein of endothelial cells and platelets. This adhesion of neutrophils was mediated by GMP140 and not by the CD18 integrin complex. By contrast, GMP140 in solution inhibited the CD18-dependent adhesion of tumor necrosis factor-alpha-activated neutrophils to plastic surfaces and resting endothelium, but not of resting neutrophils to tumor necrosis factor-alpha-activated endothelium. Thus, the binding of … Show more

“…1 sP-selectin was shown to prevent activated neutrophil adhesion to EC in vitro. 18 In the present report, no differences were observed in sP-selectin levels between normal controls and asthmatic patients. L-selectin is constitutively expressed on most leucocytes but with varying density.…”

“…The presence of P-selectin on an activated endothelial cell layer plays an essential role in the initiation of a tentative adhesive interaction between the circulating inflammatory cell and activated endothelial cell monolayer [21]. Subsequently, the enhanced expression of endothelial cell-associated PAF has been shown to cause transient adherence of neutrophils to the endothelial cells [22][23][24].…”

“…1 By contrast, intact platelets may inhibit neutrophil O 2 Ϫ generation and cytotoxicity, whereas leukocyte chemotaxis, adhesion, and O 2 Ϫ generation may be inhibited by plateletreleased NO 5 and soluble P-selectin. 6 Thus, platelets and platelet-released products may influence leukocyte function in a complex manner.…”
