Prevention and amelioration of collagen‐induced arthritis by blockade of the CD28 co‐stimulatory pathway: requirement for both B7‐1 and B7‐2

Abstract: Collagen type II-induced arthritis (CIA) is an experimental model of arthritis that has been successfully used to dissect the pathogenesis of human rheumatoid arthritis and to identify potential therapeutic targets. We have used this model to evaluate the role of T cell co-stimulation in both disease development and progression. T cell co-stimulation is provided by ligation of CD28 with either B7-1 or B7-2 present on antigen-presenting cells and can be prevented by a soluble form of CTLA-4 (CTLA-4Ig) which bin… Show more

“…In many animal models of arthritis, CD28 acts as a classic essential costimulatory protein in permitting the TCR-dependent responses to collagen that are required for initiation and progression of joint inflammation (36)(37)(38)(39). Trials with CTLA-4Ig blockade of CD28 signaling in RA have been extremely promising, but the mechanism of its therapeutic activity in humans has yet to be assessed (13,14).…”

“…CD28-deficient DBA/1 mice do not develop CIA, suggesting an absolute requirement for costimulation provided by CD28 for autoantigen-specific CD4 ϩ T cell activation and development of disease in this model (21). The prevention and amelioration of collageninduced arthritis by blockade of the CD28 costimulatory pathway have been observed (22), but not confirmed by others (23).…”

“…Studies in animal models proved its efficacy in dampening the manifestations of arthritis. Administration of CTLA-4Ig in mice at the time of the first collagen injection prevented the development of CIA (113). When treatment was delayed until arthritis had developed, CTLA-4Ig was still able to ameliorate disease.…”
