OBJECTIVES: The objective of this study was to evaluate the present status of amoebiasis in Thi-Qar Province in southern Iraq, and to determine the presence of Entamoeba histolytica and Entamoeba dispar with nested and real-time polymerase chain reaction (PCR).METHODS: Epidemiological data were obtained from the public health department of the Thi-Qar Health Office (2015-2020). Eighty stool samples were also randomly collected from patients ≤12 year of age with diarrhea at 2 hospitals between the beginning of February 2020 and the end of October 2020. These samples were selected after microscopy to identify the 18S rRNA gene in Entamoeba DNA.RESULTS: Of the 341,554 cases of intestinal parasitic infections, 38,004 (11.1%) individuals were recorded as having amoebiasis, which accounted for the highest proportion of infections in 2015 (26.1%) and the lowest in 2020 (8.1%). Amoebiasis was distributed among all age groups, with the age group of 5-14 years accounting for the highest proportion (27.3%). In molecular testing, 42 (52.5%) out of 80 samples were positive for the 18S rRNA gene (888 bp). Using nested PCR, E. histolytica (439 bp) was detected in 25 (31.3%) samples and E. dispar (174 bp) in 14 (17.5%), while using real-time PCR, E. histolytica and E. dispar were detected in 28 (35.0%) and 15 (18.8%) samples, respectively.CONCLUSIONS: Epidemiological data confirmed that amoebiasis is endemic in this province, and is not limited to certain months. Our study confirms the applicability of molecular identification to detect pathogenic and non-pathogenic Entamoeba to prescribe the appropriate drug.

Similarly, many of the researchers scored more prevalence infection of Entamoeba spp in young children's they ( Saida et 2017) in Yemen ,that they recorded high prevalence of infection (45.3 %) in age of less than ten years but not agreed with recorded lower infection rate (6.1%) in age over 41 years.…”

The current study was for determine Entamoeba histolytica in diarrheal patients by molecular investigation (nested multiplex PCR) in order obtained for an accurate diagnosis of Entamoeba histolytica that causes amoebiasis. Total stool samples (133) that collected from symptomatic patients with diarrhea according to age, gender. Then all samples undergo for a direct microscopic examination, The result revealed that 100(73.18%) were positive samples with Entamoeba spp. The results of microscopic examination revealed a higher infection rate of Entamoeba spp were in the female 83.87% compared the male 67.6% with significant difference at P<0.05 ,and recorded high infection rate in age group in (4-10)years and age group (more than 50)years 83.33%,86.66% respectively, and lower infection rate in age group (>1)year 55.55% .Showed the method nested multiplex polymerase chain reaction by using 18s rRNA small subunit gene, nested multiplex PCR for these positive samples by using specific primer for E. histolytica revealed a higher infection rate was E. histolytica 73(91.25%) (monoinfection). Entamoeba histolytica high infection rate recorded in the female 97.43% compared the male 82.35%. In addition, E. histolytica recorded the more percentage infection was in age group (less than one year) and (31-50)years was scored 3(100%), 11(100%), respectively.

Enterobius vermicularis infection is considered as one of the important causes of anaemia and malnutrition among children. This topic has recently received an increased amount of attention. The objective of this study is to evaluate the demographical, anthropometrical, nutritional, and haematological status of E. vermicularis infection among children. This study was conducted in Al Diwaniyah province, south of Iraq, for the period of October 2020 to the end of January 2021. The study included 122 children from both genders (males, n= 61, and females, n=61) and their ages ranged between 1 and 14 years. Nutritional status, body mass index (BMI), BMI percentile, and weight- for- age Z score were evaluated for some participants. In addition, samples of cellophane tape and blood were collected from all participants. The cellophane tape samples were examined microscopically for E. vermicularis detection, whereas blood samples were processed to evaluate several parameters, which were hemoglobin, serum iron, and total iron binding capacity (TIBC), ferritin, and vitamin B12. The results revealed a high infection rate (73.77%) among the studied population. The results also showed a significant correlation between gender and the occurrence of E. vermicularis infection. Males showed a higher risk of enterobiasis than females. While no association was noticed between each of age, nutritional status, and enterobiasis. Also, a significant correlation was recorded between E. vermicularis positive status and anaemia. The results showed that each of BMI, weight- for- age Z score, haemoglobin, serum iron, total iron binding capacity (TIBC), and ferritin had significantly lower values among those who were positive for E. vermicularis. Finally, the level of vitamin B12 was insignificantly decreased in children with enteroniasis. These findings indicate that more hematological changes are expected to occur in children with enterobiasis. Therefore, further consideration and investigation would be required on this topic in Iraq.
