Background: Quality and microbial safety of milk is demanding day by day as it is considered as a host for pathogenic and spoilage microorganisms. In this study, isolation and molecular characterization of shigatoxigenic O157 and non-O157 Escherichia coli in raw milk marketed in Chittagong, Bangladesh were done on 186 raw milk samples in Bangladesh. Methods: MacConkey agar was initially used to screen for the presence of E. coli and the suspected growth as evidenced by large pink colonies on MacConkey agar. Finally the organism was verified by plating through Eosin Methylene Blue (EMB) agar (a selective medium for E. coli where it produces metallic sheen) and applying standard biochemical tests for E. coli. The presence of virulent genes, Shiga-like toxin (stx1 and stx2), Intimin (eaeA), O157 antigen rfbE and Enterohemorrhagic Escherichia coli (EHEC) Hemolysin (EHEC) hlyA in the contaminating E. coli population was determined by polymerase chain reaction (PCR) run on a thermocycler (Applied Biosystem, 2720 thermal cycler, Singapore). Result: Among the raw milk samples, 33 samples were identified as E.coli positive and among the isolates, 6 (18.18%) were identified as possible EHEC O157 and rest of the isolates (81.82%) were considered as probable non EHEC O157. About, 3.23% (186 samples) EHEC O157 was isolated from raw milk samples. Then all the 33 isolates were taken under PCR assay for the identification of five virulent genes Stx1, Stx2, eaeA, rfbE and hlyA. No virulent genes were found in non- EHEC O157 isolates, but 4 stx2 (66.67%) and 1 hlyA (16.67%) gene were observed in another 4 EHEC O157 isolates out of 6, but one isolates contained the both genes and hence the prevalence of STEC was 2.15% in raw milk. Result indicated poor hygienic standard of raw milk from uncontrolled environments and the increased public health risk of those consuming raw milk from such uncontrolled sources.

Background: Cephalosporins are major antimicrobials used to treat serious infections. However, their effectiveness is being compromised by the emergence of extended spectrum beta lactamases (ESBL). The present study was aimed to characterize ESBL producing Escherichia coli from healthy and diarrheic cattle calves. Methods: A total of 52 rectal swabs were collected from healthy and diarrheic cattle calves between August 2019 to March 2020. Phenotypically E. coli were characterized based on morphology, microscopy and biochemical tests. Extended Spectrum Beta Lactamases (ESBL) E. coli initial screening was performed by using cefpodoxime, ceftazidime, cefepime and aztreonam antibiotics. The confirmation of the ESBL producing strains was conducted through ESBL phenotypic identification kit and double disc diffusion test (DDDT). Molecular characterization of ESBL E. coli was conducted by PCR amplification of blaCTX-M, bla TEM and bla AmpC genes. Plasmid profiling of the ESBL E. coli isolates was carried out. Result: Phenotypically, out of the 51 E. coli isolated, 05 (9.8%) E. coli isolates were confirmed as ESBL strains. PCR amplification revealed 01 (20%), 04 (80%) and 03 (60%) isolates positive for blaCTX-M, bla TEM and bla AmpC genes, respectively. Plasmid profiling of the isolates showed that 60% isolates were harboring plasmids of varying size and number. The plasmid number among the isolates ranged from 0-4 plasmids per isolate and plasmid size varied from 900bp to greater than 10kb. The relatively high rates of ESBL producers in diarrheic calves were found than healthy calves. The study highlights the existence of ESBL producing E. coli isolates with certain strains carrying mobile genetic elements in livestock and probable disseminators of resistance, thus imposing public health threat.

Horizontal gene transfer (HGT) in food matrices has been investigated under conditions that favor gene exchange. However, the major challenge lies in determining the specific conditions pertaining to the adapted microbial pairs associated with the food matrix. HGT is primarily responsible for enhancing the microbial repertoire for the evolution and spread of antimicrobial resistance and is a major target for controlling pathogens of public health concern in food ecosystems. In this study, we investigated Salmonella Heidelberg (SH) and Escherichia coli (EC) regarding gene exchange under conditions mimicking the industrial environment, with the coproducts whey (SL) and chicken juice (CJ). The S. Heidelberg strain was characterized by antibiotic susceptibility standards and PCR to detect the blaTEM gene. A concentration of 0.39 mg/mL was determined to evaluate the anti-conjugation activity of nanostructured lipid nanocarriers (NLCs) of essential oils to mitigate β-lactam resistance gene transfer. The results showed that the addition of these coproducts promoted an increase of more than 3.5 (whey) and 2.5 (chicken juice) orders of magnitude in the conjugation process (p < 0.01), and NLCs of sage essential oil significantly reduced the conjugation frequency (CF) by 74.90, 90.6, and 124.4 times when compared to the transfers in the absence of coproducts and the presence of SL and CJ, respectively. For NLCs from olibanum essential oil, the decrease was 4.46-fold for conjugations without inhibitors and 3.12- and 11.3-fold in the presence of SL and CJ. NLCs associated with sage and olibanum essential oils effectively control the transfer of antibiotic resistance genes and are a promising alternative for use at industrial levels.
