Presumptive TRP channel CED-11 promotes cell volume decrease and facilitates degradation of apoptotic cells in Caenorhabditis elegans

Abstract: Apoptotic cells undergo a series of morphological changes. These changes are dependent on caspase cleavage of downstream targets, but which targets are significant and how they facilitate the death process are not well understood. In Caenorhabditis elegans an increase in the refractility of the dying cell is a hallmark morphological change that is caspase dependent. We identify a presumptive transient receptor potential (TRP) cation channel, CED-11, that acts in the dying cell to promote the increase in apopto… Show more

“…It has been previously noted that several species express TRPM-like channels which cluster independent of, or differ greatly from, other chordate and/or insect TRPMs [28,52,81]. In accordance with this, the consensus TRPM phylogeny shows a group of protostome TRPMs located basally, near choanoflagellate TRPMs (figure 5b, green).…”

“…The function of TRPS, however, remains mostly mysterious. Only a single member has been functionally characterized: C. elegans ced-11 , which encodes a Ca 2+ channel which participates in apoptosis [110].…”

“…This decrease is likely induced by the opening of potassium and chloride channels in the plasma membrane, causing an efflux of potassium and chloride ions followed by water [ 4 ]. Blocking this shrinkage in apoptotic cells in Caenorhabditis elegans embryos compromises their ability to die, which suggests that cell shrinkage facilitates the execution of apoptosis [ 5 ].…”
