Present and future scope of recombinant parathyroid hormone therapy in orthopaedics

Abstract: Parathyroid Hormone (PTH) has a significant role in calcium metabolism. Its intermittent administration has an anabolic effect on bone mineralization. Teriparatide (PTH 1-34), a recombinant form of parathyroid hormone, is useful in the treatment of osteoporosis, fracture healing, non-union, stress fracture, augmentation of implant fixation with bone, and chondroprotection in osteoarthritis. The present review article will elaborate on the potential approved uses of recombinant PTH in orthopedics and its evolvi… Show more

“…Strategies to develop disease-modifying-treatments and newly established KOA medications have attracted much attention, which would reduce the costs and benefit patients. PTH (1–34) was used to treat OP, improve healing of the bone fracture widely [ 24 ], and emerge as a promising agent for KOA treatment. Therefore, PTH (1–34) might be a medication for KOA but the potential mechanism of its pharmacological effects remains to be explored.…”

“… 7 On the contrary, anti-osteoporotic agents like teriparatide have been tried to enhance fracture healing due to its anabolic effect. 8 Following an acute fracture, the optimal timing of starting the osteoporosis medications especially for antiresorptive agents like bisphosphonate remains an important concern. The purpose of this review is to assess the current literature on the effect of timing of initiation of different osteoporosis medications following an incident fracture.…”
