Presenilin-deficient neurons and astrocytes display normal mitochondrial phenotypes

Abstract: Presenilins 1 and 2 (PS1 and PS2) are predominantly known as the catalytic subunits of the γ-secretase complex which generates the amyloid-β (Aβ) peptide, the major constituent of the senile plaques found in the brain of Alzheimer’s disease (AD) patients. Apart from their role in γ-secretase activity, a growing number of cellular functions have been recently attributed to PSs. They are involved in synaptic transmission, endo-lysosomal function and calcium homeostasis. PSs were also found to be enriched in mito… Show more

“…Primary cultures of neurons were obtained from postnatal day 0 pups as previously described [40, 41], with some modifications. Briefly, brain tissues (containing cortices and hippocampi) were isolated by dissection in ice-cold HBSS/0.2% glucose and meninges were removed.…”

“…The sequence encoding human APP695 carrying 3 FAD mutations (Swedish [K670N/M671L], Florida [I716V], and London [V717I]) was cloned into the pLVX-CMV bicistronic retroviral vector, to produce the hAPP3xmut plasmid. Lentiviral particles were generated as previously described [41, 42]. Briefly, HEK293T cells were transfected with the hAPP3xmut, pMD2.G (Addgene #12259), and pCMV-dR8.2 (Addgene #12263) plasmids.…”
