Presence of Multiple Functional Polyadenylation Signals and a Single Nucleotide Polymorphism in the 3′ Untranslated Region of the Human Serotonin Transporter Gene

Abstract: Abstract:The human serotonin transporter (hSERT) gene is a candidate for involvement in the aetiology of affective disorders. In humans, multiple transcripts of the gene have been detected by northern blot analysis of brain and other tissues. We performed 3Ј rapid amplification of cDNA ends to identify the common sites of polyadenylation in hSERT mRNA from human JAR cells and whole blood. Two major polyadenylation sites were identified: one 567 bp downstream of the stop codon, consistent with the usage of the … Show more

“…26,31,40 Four investigated other genes 37,43,64,65 and three investigated polymorphisms other than the 5-HTTLPR and the intron 2 VNTR. 36,45,55 Six papers studied a specific clinical feature of BPD rather than the illness itself. 11,[28][29][30]72,73 Finally, two did not provide enough data to be included in the meta-analyses.…”

“…The SERT mRNA occurs in two alternative polyadenylation forms that differ by the presence or absence of a conserved sequence element in the 3 0 untranslated region (Battersby et al, 1999;Gyawali et al, 2010). Increased expression of SERT mRNA containing the distal polyadenylation sequence appears associated with decreased anxiety.…”

“…22,23 Transcriptional activity is regulated by several positive and negative regulatory elements within the SERT promoter region [23][24][25][26] and by a 17 bp variable number tandem repeat (VNTR) element in intron 2. 27 Differential splicing 28,29 and use of two 3 0 polyadenylation sites 30 may also contribute the regulation of SERT expression. 31 Heils et al 25,32 first described functional variants of a repetitive sequence in the SERT linked polymorphic region (SERTLPR) located 1.2 kb upstream of the transcription start site.…”
