Preparative separation of liquiritigenin and glycyrrhetic acid from Glycyrrhiza uralensis Fisch using hydrolytic extraction combined with high‐speed countercurrent chromatography

Abstract: The objective of this paper was to develop a preparative method for the isolation and purification of liquiritigenin and glycyrrhetic acid from Glycyrrhiza uralensis Fisch using hydrolytic extraction combined with high‐speed countercurrent chromatography (HSCCC). Liquiritigenin and glycyrrhetic acid were well hydrolyzed from liquiritin and glycyrrhizic acid by hydrochloric acid, respectively. The optimal extraction conditions were obtained by single‐factor and orthogonal experiments, which were 100% ethanol, 1… Show more

“…The main bioactive flavonoid compounds in RG, LTG, and ISL were identified and isolated from the crude extract of Glycyrrhiza uralensis [ 165 , 166 ]. ISL demonstrates antioxidant, anti-inflammatory, antitumor, and hepatoprotective activities.…”

“…GLR is commonly isolated from Glycyrrhizae Radix in the form of a monoammonium salt (C 42 H 61 O 16 _NH 4 ), but occasionally studies are performed with the magnesium salt of 18β-glycyrrhizic acid (magnesium isoglycyrrhizinate) (Cao et al, 2019;Tan et al, 2018;Zou et al, 2018). Specific methods have been developed to monitor the extraction process and for GLR dosages in biological media (Wang, Shan, & Lü, 2020;Yu et al, 2017;Zeng et al, 2020).…”

“…In addition to HMG proteins, GLR has also been shown to bind to other proteins such as serum albumin Wang, Shan, & Lü, 2020) and to recombinant HIV-1 reverse transcriptase to inhibit the protein phosphorylation (Harada et al, 1998). Binding of GA to the active site of lactate dehydrogenase from Plasmodium falciparum (malaria parasite) has been proposed based on molecular modeling, mimicking the binding of chloroquine to the same site and possibly explaining the modest antimalarial activity of GLR (Kalani et al, 2013).…”

“…Phenols comprise a wide variety of compounds such as polyphenols, flavonoids, condensed tannins. Phenolic compounds are mostly hydrophilic, while the flavonoids are mostly hydrophobic [30]. However, due to their structures, flavonoids such as quercetin, kaempferol and catechin were easily extracted by hydromethanolic solvents.…”
