Unstable linkage between genetic markers in transformation. J. Bacteriol. 89:1314-1321. 1965.-A new type of association of genetic markers in a transformation system has been discovered. Linkage between certain markers is detectable only under particular conditions of deoxyribonucleic acid (DNA) extraction and is sensitive to dilution of the DNA. The data suggest that this unstable association occurs between markers which are on the same molecule but separated by a molecular distance sufficiently great for linkage to be preserved only by mild DNA extraction procedures and at high concentrations of DNA. Linkage can be stabilized by dilution in the presence of a carrier DNA. By use of these unstable linkages, the genetic map of Bacillus subtilis has been extended in both directions from the aromatic gene cluster.

Priming by homopolynucleotides directed the incorporation of the complementary nucleotides. There was competition between the various primers suggesting that a single enzyme is involved in the reactions studied. Poly fluorouridylate was an effective inhibitor of both the DNAand polyribonucleotide-primed reactions but actinomycin D, which inhibits the reaction primed by DNA, had no appreciable effect on the polyribonucleotide-primed reaction.

A study has been made of the levels of 6-methylaminopurine and ymethylcytosine in the DNA of B d c i h subtilis during thymine deprivation. While DNA synthesis was inhibited by thymine deprivation, DNA methylation continued. Base analysis indicated that this aberrant methylation involved an increase solely in the amount of 5-methylcytosine. These aberrant 5-methylcytosine residues were removed from the DNA during continued growth of bacteria in medium lacking thymine. In contrast, 5-methylcytosine residues synthesized during normal growth were relatively unaffected by thymine deprivation. The results are interpreted to indicate that the extensive DNA damage which occurs during thymine deprivation is due in part to exonuclease digestion of regions of DNA containing aberrant 5-methylcytosine residues.
