Preparation of novel trivalent vaccine against enterotoxigenic Escherichia coli for preventing newborn piglet diarrhea

Abstract: OBJECTIVE To develop a trivalent genetically engineered inactivated Escherichia coli vaccine (K88ac-3STa-LTB) that neutralizes the STa toxin by targeting fimbriae and entertoxins for the treatment of enterotoxigenic E coli. ANIMALS 18- to 22-g mice, rabbits, pregnant sows. PROCEDURES Using PCR, the K88ac gene and LTB gene were cloned separately from the template C83902 plasmid. At the same time, the 3 STa mutant genes were also amplified by using the gene-directed mutation technology. Immune protection exp… Show more

“…A significant observation from our dataset was the reduction in the Escherichia-Shigella population within the Phy group. This is particularly relevant because E. coli, a member of this group, is primarily associated with postweaning diarrhoea (Xu et al 2023). This condition poses a considerable economic challenge for pig farmers due to its association with reduced weight gain, increased mortality, and higher susceptibility to secondary infections.…”

“…However, currently, there are challenges in strain expression level for preparing the bacterial antigen protein itself along with obtaining sufficient soluble protein and facing high purification costs. These obstacles have impeded its immune effectiveness and hindered its application in clinical practice thus far [ 24 , 25 , 26 ]. Hence, there exists an urgent necessity for an efficacious vaccine solution to prevent and control ETEC.…”

“…Enterotoxigenic Escherichia coli (ETEC) strains are gram-negative enteric pathogens that are the most common cause of diarrhoea in humans and animals (Xu et al 2023 ). ETEC is also a major cause of diarrhoea in neonatal piglets and weaned piglets, resulting in significant economic losses to farmers (losses due to mortality, morbidity, reduced growth rate, and medication costs).…”
