Preparation of hydroperoxy and hydroxy derivatives of rat liver phosphatidylcholine and phosphatidylethanolamine

Abstract: A convenient method for the preparation of hydroperoxy and hydroxy derivatives of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) is described. PC and PE obtained from rat liver were oxidized with singlet oxygen by using methylene blue as the photosensitizer, and their hydroperoxides were isolated with the aid of reverse phase liquid chromatography. The hydroxy derivatives were obtained by reducing the hydroperoxides with sodium borohydride. The results of gas chromatography mass spectrometry reveal… Show more

“…Then it was determined at 235 nm by the HPLC method using the Shimadzu system, C8 Nova Pak column and methanol-water mixture (96:4 v/v) as a mobile phase. The amount of peroxides was calculated from the standard curve prepared with PC-OOH according to the method described by TERAO et al (1985).…”

“…Hydroperoxides of cholesteryl esters, triacylglycerols and phospholipids were monitored by HPLC as described elsewhere (21,22). Formation of carbonyls was monitored according to Levine et al (23) and thiol (-SH) content was determined according to Elman (24).…”

“…Phosphatidylcholine hydroperoxide (PC-OOH) was prepared from EYPC by the photoirradiation method ( 9 ). A sample of 100 mg of EYPC dissolved in 5 mL of methanol containing 0.1 m M methylene blue was illuminated with a 30-W tungsten projection lamp at room temperature for 12 h. After the methylene blue was removed by silica gel column chromatography, a crude PC-OOH sample was obtained.…”
