Preparation of Engineered Extracellular Vesicles Derived from Human Umbilical Cord Mesenchymal Stem Cells with Ultrasonication for Skin Rejuvenation

Abstract: Extracellular vesicles (EVs) are lipid-bilayer-enclosed vesicles of submicron size that are secreted by various cells. As mediators of intercellular communication, EVs can alter the physiological state of recipient cells by delivering encapsulated proteins and nucleic acids. Incontestably, growing evidence has shown important biological roles and the clinical relevance of EVs. The use of stem cell-derived EVs as a cell-free therapeutic modality for skin treatment has emerged as a promising application in derma… Show more

“…EVs were isolated by differential centrifugation as previously described. 29 Briefly, the culture medium was centrifuged at 300×g for 10 min to remove cells and at 3000×g and 10,000×g to remove cell debris. Then, EVs were pelleted by ultracentrifugation at 120,000×g for 90 min and afterward washed by PBS.…”

“…For instance, nitrogen cavitation was used to instantly disrupt neutrophil-like HL60 cells leading to a 16-fold increase in the formation of nanosized membrane vesicles, which were then loaded with an anti-inflammation drug to treat acute lung inflammation/injury and sepsis induced by LPS 249 . Ultrasonication of MSCs for 1 min also improved the EV yield, which exhibited ∼20-fold higher and ∼100-fold faster production than the natural secretion 250 . A recent study described a detailed method to fabricate an origami-paper-based device that integrated the ion concentration polarization technique for effective enrichment and isolation of EVs in a simple manner 251 .…”

“…An alternative way of producing EVMs from MSCs is via ultrasonication. Wang et al (2019) used 1 min ultrasonication for delivery of shearing force to intact MSCs, followed by centrifugation to produce EVMs [ 63 ] ( Figure 2 B). With the same number of cells, ~20-to-100-fold more EVMs can be generated than naturally secreted EVs [ 29 , 62 ].…”
