Preparation of Amino Alcohols Condensed with Carbohydrates: Evaluation of Cytotoxicity and Inhibitory Effect on NO Production

Abstract: This work reports the preparation of several amino alcohols condensed with D-arabinose, D-glucose, and D-galactose derivatives. These compounds were evaluated in vitro for their cytotoxicity and ability to decrease nitric oxide production in J774A.1 cells. Arabinofuranoside derivatives 5a, 5b and 5c showed a significant inhibition of nitric oxide production (>80% at 5 lg ⁄ mL), while the galactopyranoside derivative 8d showed a notable nitric oxide inhibitory activity (126% at 0.5 lg ⁄ mL).

“…Lipophilic drugs pass more easily through biological membranes than hydrophilic drugs, increasing their intracellular concentration and modulating signaling pathways, transcription factor activity, and gene expression (10,15). The genistein derivatives synthesized in this work showed different effects on NO, IL-12, and TNF-a production.…”

“…The nitrites production was quantified by comparing with a standard curve, made with different concentrations of NaNO 2 . The reading was measured in the microplate reader (Spectramax 190; Molecular Devices) at 540 nm wavelength (14,15).…”

“…A correlation between hydrophobic character, membrane permeability and biological activity is frequently observed [18,19]. In this work, a novel analog of genistein, the 7-O-tetradecanoyl-genistein (TDG), was developed using esterification in vitro to bypass the in vivo processing, which favors biological activity and increased lipid solubility.…”

“…The present work describes the synthesis of novel anthraquinone derivatives conjugated with lipidic amino alcohols. This class of compounds has been shown to possess immune modulation activity and antiparasitic and antimicrobial properties (11)(12)(13)(14)(15)(16). The new compounds were evaluated for their effects on NO, IL-1b and TNF-a production in LPS/IFN-c-stimulated RAW264.7 macrophages.…”
