Preparation of algal-oligosaccharide mixtures by bacterial agarases and their antioxidative properties

Abstract: Algal-oligosaccharide-lysates (AOL), derived from six agars and four algal polysaccharide extracts (APE), were treated with 100-500 activity units (AU) of MA103-agarases or MAEF108-agarases, and their antioxidative properties evaluated. Soluble total polyphenols (TP) were between 462.2 ± 1.6 gallic acid equivalents (GAE, mg/mL) and 70.6 ± 17.4 GAE. The DPPH radical scavenging capacity of all AOL went from 68.3 ± 0.7% to 0.5 ± 0.1%. The ferrous ion chelating capacity of all AOL went from 93.1 ± 0.2% to 21.7 ± 0… Show more

“…From the results, the decrease in DPPH-radical indicated the radical-scavenging effect of porphyrans. It is obvious that the scavenging effects obeyed the order of P 3 > P 2 > P 1 > P. Wu and Pan [6] had demonstrated the similar result. They attributed the increasing effect on scavenging DPPH-radical of agar polysaccharide extracts from Porphyra dontate to increased oligosaccharide production caused by increased agarase activity units.…”

“…Structurally, it has a linear backbone of alternating 3-linked ␤-d-galactosyl units and 4-linked ␣-l-galactosyl 6-sulfate or 3,6-anhydro-␣-l-galactosyl units [1]. Bioasssays have shown that porphyran could stimulate immune response [2,3], scavenging active oxygen radicals in vitro [4][5][6], increase antioxidant enzymes activity and antioxidant capacity in aging mice [7]. However, porphyran is a high molecular weight polysaccharide featuring high viscosity, which may limit its pharmaceutical application.…”

“…Immune modulating activity of HCP Expression of iNOS and COX-2 in macrophages is regulated by transcriptional activation during immune responses and inflammation [12,50,51]. Therefore, RT-PCR analysis was performed to evaluate the effects of purified HCP on macrophage activation by measuring the expression levels of iNOS and COX-2 mRNA from RAW264.7 cells.…”

“…Compared with the 100 ppm Vitamin C, 20% Chlorella hydrolysates had higher TEAC and reducing power values ( P < 0.05). These phenomena suggested that, not only the hydrolyzed soluble proteins and peptides, but the released polysaccharides also have iron chelating and free radical scavenging abilities (Wu and Pan 2004; Yin and others 2007). Chlorella contains many dietary antioxidants that could eventually be responsible for some of its functional activities.…”
