Preparation of a protein‐free total brain white matter lipid fraction: Characterization of liposomes

Abstract: A method of preparing a total lipid extract (TLE), free of protein, by extracting brain white matter with tetrahydrofuran is presented. The optimal conditions of extraction were found to be 50 ml of THF per gram of lyophilized tissue, though fresh tissue can also be used if larger volumes of solvent are employed. The method allowed, in a short time and in a single step, a yield of TLE of 50% on a dry weight basis. Its analytical characterization revealed a qualitative and quantitative composition very similar … Show more

“…This polar ether-based 3DISCO protocol has achieved extremely rapid dehydration due to its high permeable kinetics and has provided fully cleared organ samples after RI matching. Because THF has significant lipophilicity (Díaz et al 1992), this protocol may accompany highly effective delipidation. THF-solvated tissues are more slightly shrunk than EtOH-solvated ones, probably because THF lacks donating hydrogen.…”

“…We examined two more complicated model systems: total brain extract lipids (TBE-lipids) and a ternary system consisting of DOPC, 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and cholesterol at a 1:2:1 ratio. TBE-lipids have been widely used as models of the plasma membrane and have been used in studies of IAPP-membrane interactions (28,(39)(40)(41). They are considerably more complex than standard one-or two-component model membrane systems.…”

“…1) characterized by an EY value of 94% and an EC value of 14 l/ mol. The EC value was much higher than that expected for unilamellar liposomes [26]. This finding may be due to two different factors: i) the precipitation of the drug molecule as sulfate salt (used for the generation of the acid pH gradient) in aqueous compartments of unilamellar liposomes and/or ii) the interaction of gemcitabine with the molecules of phospholipids [27].…”
