Preparation of a population of macrophages from human third trimester decidua

Abstract: Human decidua in the third trimester of pregnancy contains at least four different cell types (stromal cells, macrophages, T-lymphocytes and granulocytes). The contributions of these cells to the overall function of the tissue are not known, although it is anticipated that both stromal cells and macrophages will produce the prostaglandins involved in labour. In addition, the decidua produces cytokines, but the cellular source has not been identified. As a first stage to addressing these questions, we have deve… Show more

“…This latter finding highlights the complexity of the decidua and its specialized biology, which to date has not been exploited for therapeutic intervention. Several studies have described methods that yield highly purified decidual cell preparations [17][18][19] and have been utilized to investigate immune [17,20,21] and endocrine [18] aspects of decidual biology but, hitherto, not their electrophysiology.…”

“…This may explain why decidua explants secrete slightly less secretory leukocyte protease inhibitor compared with chorio-decidua, unless there is a gradient of secretion with highest outputs from decidua adjacent to the chorion. Term decidua is heterogeneous, consisting of stromal cells (53%), macrophages (19%), granulocytes (16%) and T-cells (8%) (Adeleye et al 1996). Hence the stromal cells, macrophages and granulocytes, which are capable of secretory leukocyte protease inhibitor secretion in other sites (Ohlsson et al 1996), are potential sources.…”

“…Macrophages within the uteroplacental environment are an important source of bioactive mediators including prostaglandins and cytokines. Placental and decidual macrophages express COX-2 and produce PGE 2 in response to LPS or the pro-inflammatory cytokine IL-1β [ 25 29 ]. No studies to date have examined the effects of environmental toxicants, such as MEHP, on inducible COX-2 expression or prostaglandin secretion in macrophages from the utero-placental unit.…”
