Preparation of 99 mTc-MAG3: the effect on radiochemical purity of using sodium chloride injection from plastic ampoules that have been exposed to light

Abstract: Preparing 99m Tc-MAG3 using SCI from plastic ampoules that have been exposed to light causes reduced RCP. Exposure of plastic ampoules to light causes leaching of many compounds into the solution. An unknown leached compound destabilizes the 99m Tc-tartrate intermediate complex.

“…It is possible that the effect of light might be different with ampoules from other manufacturers. This is illustrated by our previous work [1] in which we demonstrated that the effect on radiochemical purity was more pronounced with Steripak ampoules (Norton, Cheshire, UK).…”

“…In previous work on 99m Tc mertiatide, we demonstrated that when plastic ampoules containing sodium chloride injection are exposed to light, many contaminants are formed in the solution but we were unable to identify them or isolate which were responsible for the reduction in radiochemical purity [1]. This study shows that these contaminants also reduce the degree to which exametazime is labelled and slow the rate at which 99m Tc labels albumin nanocolloid and macrosalb.…”

“…It has been noted that saline ampoules exposed to light can adversely affect the formation of the desired complex; curiously, this appears to affect the formation of 99m Tc tartrate rather than the transchelation step. 33 It has also been reported that the duration of the boiling step is critical. 34 There was a report that suggested that the quality of sestamibi kits had declined over the years, as evidenced by deterioration in the quality of dual-phase parathyroid scintigraphic studies.…”
