Preparation and quality by design assisted (Qb-d) optimization of bioceramic loaded microspheres for periodontal delivery of doxycycline hyclate

Abstract: PLGA (Lactic- co-glycolic acid) coated chitosan microspheres loaded with hydroxyapatite and doxycycline hyclate complex were developed in the present study for periodontal delivery. A modified single emulsion method was adopted for the development of microspheres. Formulation was optimized on the basis of particle size, drug loading and encapsulation efficiency with the central composite design using 2 3 factorial design. Microspheres were optimized and electron microscopy revealed their… Show more

“…The developed complex was characterized on the basis of surface morphology by SEM, so as to confirm the development of the complex. 38 Further, in order to calculate the amount of drug adhered to the HAP surface, a weighed amount of the HAP-Doxy dried composite was added to 10 mL of water. The suspension was then filtered with the help of Whatman filter paper, and the unadsorbed drug in solution was determined with a UV−visible spectrophotometer (UV-1601, Shimadzu, Japan) at 271.5 nm.…”

“…The high and low levels of the given factors were coded by +1 and −1, respectively, which were used to determine the relative impact of the independent variables compared to their coefficients. 42 3.3.1 Effect of the independent variables on the size of the particles. Particle size (PS) is considered to be a critical parameter in the CQAs in the fabrication of MPPs, given that it facilitates the penetration and release of the therapeutic drugs.…”

“…In addition, the introduction of antibiotics or drugs into bionic bone scaffolds and the use of bioceramics and scaffolds to control their release can increase the antibacterial activity. Lactic-co-glycolic acid (PLGA)-coated chitosan microspheres loaded with HA and doxycycline hyclate complexes have been developed for periodontal delivery [ 298 ]. Endodontic sealers that incorporate novel, highly loaded antimicrobial drug-silica coassembled particles (DSPs) show great antimicrobial activity [ 299 ].…”
