An environmentally-friendly wood adhesive was developed by blending konjac glucomannan (KGM), chitosan (CH), and polyvinyl alcohol (PVA) together. The viscosity of the KGM-CH-PVA (KCP) blend adhesive was determined, and the morphology of the film was observed using scanning electron microscopy (SEM). The KCP blend adhesive was applied to plywood during the manufacturing process, and the effects of the KGM, CH, and PVA contents on the bond strength was investigated. Results showed that KGM greatly increased the viscosity of the KCP blend adhesive, whereas the addition of PVA decreased the viscosity in the test range. The SEM observations showed that the KCP blend adhesive was homogeneous. The bond strength of the plywood that was treated with KCP blend adhesive increased with increasing KGM and CH concentrations, and desirable performance could be obtained with a total solids content of 4.6%. The KCP blend adhesive with 2.0% KGM, 2.0% CH, and 0.6% PVA exhibited a comparable bond strength with phenol formaldehyde. Findings suggest that the KCP blend adhesive can be used as a wood adhesive with all raw materials, having the advantage of being environmentally friendly.

A b s t r a c t. A comprehensive understanding of the uprooting failure mechanism will likely require the accurate characterization of the mechanical properties of cotton stalk. Uprooting failure includes a fractured cotton stalk and peeled phloem sliding along the xylem. The modulus of elasticity of cotton stalk and its tissues (xylem and phloem) were measured using three different modes (tensile, compression and bending), and the reasons for the fractured cotton stalk and the peeled phloem sliding along the xylem were analysed from the perspective of composite mechanics. The results showed that the cotton stalk radially conforms to the properties of the composite with transverse anisotropy. The axial modulus of elasticity was significantly larger than the radial modulus of elasticity (axial modulus of elasticity: cotton stalk is 3181.79 MPa, xylem is 1093.91 MPa, phloem is 249.89 MPa, radial modulus of elasticity: is 91.04 MPa, xylem is 83.77 MPa, phloem is 77.01 MPa). Xylem is the backbone of the stalk that provides 96% of its compressive strength. The direct cause of fractured cotton stalk originated from the load force that exceeded its intrinsic compressive strength. Peeled phloem sliding along the xylem was related for the most part to the different radial modulus of elasticity of the xylem and phloem, and the weak cohesion between these two tissues. Somatic Embryogenesis and Indirect In Vitro Plant Regeneration in Amorphophallus konjac K. Koch by One-Step Seedling Formation

Konjac (Amorphophallus konjac K. Koch) is a well-known tuberous vegetable belonging to the important medicinal family Araceae, and the plant grows from an underground tuber. Here, we used a “one-step seedling regeneration” tissue culture system to improve the plantlet regeneration efficiency of konjac using young leaves as an explant source. In the current study, we used several sterilization methods for tuber sterilization. Moreover, various plant growth regulator combinations were applied to achieve efficient somatic embryogenesis and plantlet regeneration. Our results showed that the optimal tuber sterilization was method C (75% alcohol for 15 s + 0.1% HgCl2 for 15 min + washing by double-sterilized water three times). Three types of embryogenic calli were induced on full-strength Murashige and Skoog (MS) basal medium supplemented with 0.5 mg/L of 6-benzylaminopurine (6-BA), 0.5 mg/L of naphthaleneacetic acid (NAA), 1.0 mg/L of 2,4-dichlorophenoxyacetic acid (2,4-D), and 30 g/L of sucrose. Of the three types of embryogenic calli, only type Ⅲ further regenerated plantlets, with a callus induction rate of 55.73% and a seedling induction rate of 92.73%. This suggests that the addition of the above hormones gives the optimal callus induction. The proliferation rate achieved was 38% on the MS basal medium containing 1.0 mg/L of 6-BA, 1.0 mg/L of indolebutyric acid (IBA), 0.2 mg/L of kinetin (KT), and 50 g/L of sucrose. The one-step seedling formation achieved in MS medium contained 2.0 mg/L of 6-BA, 0.5 mg/L of NAA, 0.1 mg/L of gibberellic acid (GA3), and 30 g/L of sucrose, and the number of regenerated shoots per explants was 6 ± 2. Therefore, we establish a one-step seedling regeneration system through indirect plant regeneration, which shortens the time for konjac in vitro regeneration, significantly increased the micropropagation efficiency, and decreased the cost of the konjac tissue culture.
