Premature Expression of the Winged Helix Transcription Factor HFH-11B in Regenerating Mouse Liver Accelerates Hepatocyte Entry into S Phase

Abstract: Two-thirds partial hepatectomy (PH) induces differentiated cells in the liver remnant to proliferate and regenerate to its original size. The proliferation-specific HNF-3/fork head homolog-11B protein (HFH-11B; also known as Trident and Win) is a family member of the winged helix/fork head transcription factors and in regenerating liver its expression is reactivated prior to hepatocyte entry into DNA replication (S phase). To examine whether HFH-11B regulates hepatocyte proliferation during liver regeneration,… Show more

“…One possible explanation for this discrepancy is that Foxm1 transgenic protein is transcriptionally inactive in quiescent lungs. It was previously demonstrated (Ye et al, 1999) that in quiescent cells transgenic Foxm1 protein localizes in cytoplasm and needs an activation signal (injury-induced proliferative stimuli) to be translocated Figure 3 Gene expression profile in Rosa26-Foxm1 transgenic lungs after 3-methylcholanthrene (MCA)/butylated hydroxytoluene (BHT) treatment. Total RNA was prepared from left lung lobes of untreated or MCA/BHT-treated wild-type (WT) and Rosa26-Foxm1 mice.…”

“…Ten to twenty micrograms of total RNA were hybridized with [␣-32 P]UTP-labeled antisense RNA probes synthesized from the appropriate plasmid templates and digested with RNase ONE (Promega, Madison, WI) as described previously. 17 Syntheses for antisense mouse HNF-6, HNF-4␣, C/EBP␣, FoxA2 (also known as HNF-3␤), insulinlike growth factor 1, and cyclophilin riboprobes have been described. 18 The mouse CYP7A1 ribonuclease protection assay (RPA) probe plasmid was obtained via reverse transcription polymerase chain reaction of mouse liver RNA (using primers 5Ј-GCA AGG ATC CTA CTT CTG CGA AGG CAT TTG G-3Ј and 5Ј-GCC GGA ATT CAA ACA TCA CTC GGT AGC AGA A-3Ј) and cloned into the pBluescript SK(ϩ) template, and an antisense RNA probe was synthesized from a BamHI digested template using T7 RNA polymerase.…”

“…10 Determination of the number of hepatocytes and mesenchymal cells undergoing DNA synthesis was performed by monoclonal antibody detection of BrdU incorporation (Roche Molecular Biochemicals, Indianapolis, IN) of injured liver using microwave retrieval to enhance antigenic activity as described previously. 20,21,24 In each liver, we counted the number of BrdU-positive nuclei per 5 ϫ200 viewing fields of liver sections and SD was calculated for 0, 1, 3, and 7 days after CCl 4 treatment using 3 mice as described previously. 24 Ribonuclease Protection Assay.…”

“…20 The mice were killed by CO 2 asphyxiation; liver tissue was used to prepare total RNA and protein cell extract, or livers were fixed overnight in 4% paraformaldehyde at 4°C and then paraffin embedded as described previously. 21 National Institutes of Health Guidelines for Animal Care were followed during CCl 4 mouse liver injury experiments.…”
