Preliminary experience with tigecycline treatment for severe infection in children

Abstract: Although some patients benefited from tigecycline, the efficacy and safety of tigecycline should not be overvalued. Additional data from randomized controlled trials are required to assess the administration of tigecycline. What is Known: • Severe infection is a primary cause of mortality in pediatric patients and its treatment is facing challenges from an increasing number of multidrug-resistant (MDR) pathogens. • Tigecycline has an expanded spectrum of antibacterial activity. • Several case reports have indi… Show more

“…Studies on the efficacy of tigecycline in children are not prospective due to safety concerns but contain data from limited cases in which it was used as salvage therapy. [10][11][12][13][14] The FDA and European Medicines Agency have issued warnings regarding the use of tigecycline in children over 8 years of age. Acinetobacter species to carbapenem decreased to 6.2% from 2010 to 2017, and that to tigecycline was >80%.…”

“…It is seen that the use of tigecycline in children is generally preferred in severe infections such as bloodstream infection, meningitis, intraabdominal infection, and nosocomial pneumonia. 7,[12][13][14] In our study, tigecycline was generally used for indications such as sepsis, febrile neutropenia, and skin and soft tissue infections.…”

“…The patient in this case suffered from intolerable diarrhea leading to the discontinuation of tigecycline combination therapy. The result might also be attributed to other factors, such as a worse clinical condition and concomitant drugs ( 26 ). Therefore, additional data from controlled clinical studies are required to assess the safety of tigecycline, and particular caution should be given to off-label use of tigecycline, especially in pediatric patients.…”
