Preliminary Data on Some Behavioral Changes Induced by Short-Term Intraperitoneal Oxytocin Administration in Aged Rats

Abstract: Results: Increased mobility and decreased anxiety behaviors were reported for the aged intraperitoneal OT-treated animals, as compared with controls, during FST and OFT, and respectively FST, EPM, and OFT. Also, decreased depressive-like behaviors were observed in the same animal group during FST and ST. Moreover, a decrease in anxiolytic behavior was observed as exposed to stressful stimuli (such as grooming behavior in OFT, and forced grooming behavior in ST), and as exposed to social stimuli (such as groomi… Show more

“…It is well known that SD induces a clear anxiogenic effect (Albrechet-Souza et al, 2017;Macedo et al, 2018), which is completely blocked by OXT administration. Although data are controversial (for example, Grippo et al, 2012), many studies indicate that OXT exerts an anxiolytic effect in both sexes and even in aged rats (Balmus et al, 2018;Waldherr and Neumann, 2007). However, we did not observe any OXT effect on non-stressed mice.…”

“…OXT treatment began at PND90 when rats reached an average weight of 150 g. OXT (Sigma-Aldrich Co., LLC., Darmstadt, Germany) was intraperitoneally injected in the IP OXT-VPA group in a dose of 10 mg/kg/bodyweight for 8 days consecutively. This dosage was established based on previous published work of our group [ 54 , 55 ] and others [ 56 ]. Behavioral tests were initiated on the 6th day of OXT treatment and lasted three days during which the OXT administration continued.…”

“…Y-Maze Task . For assessing short-term memory modifications, we used the reliable Y-maze task, as described in a previous study [52]. The spontaneous alternation percentage, defined by the consecutive entries into all three arms of the maze, was analyzed as the behavioral marker to reflect spatial working memory.…”

“…Elevated Plus Maze Task (EPM) . Anxiety-like behavior was assessed in the EPM paradigm which was also described in our previous study from 2018 [52]. Rats were placed at the juncture of the open and closed arms, and the amount of time spent on the open arms was recorded during a 5 min test.…”
