Dysfunction and loss of neurons are the major characteristics of CNS disorders that include stroke, multiple sclerosis, and Alzheimer's disease. Activation of the Toll-like receptor 7 by extra-cellular micro-RNA let-7, a highly expressed microRNA in the CNS, induces neuronal cell death. Let-7 released from injured neurons and immune cells acts on neighboring cells, exacerbating CNS damage.Here we show that a synthetic peptide analogous to the mammalian PreImplantation factor (PIF) secreted by developing embryos and which is present in the maternal circulation during pregnancy inhibits the biogenesis of let-7 in both neuronal and immune cells of the mouse. The synthetic peptide, sPIF, destabilizes KH-type splicing regulatory protein (KSRP), a key microRNA-processing protein, in a Toll-like receptor 4 (TLR4)-dependent manner, leading to decreased production of let-7. Furthermore, s.c. administration of sPIF into neonatal rats following hypoxic-ischemic brain injury robustly rescued cortical volume and number of neurons and decreased the detrimental glial response, as is consistent with diminished levels of KSRP and let-7 in sPIF-treated brains. Our results reveal a previously unexpected mechanism of action of PIF and underscore the potential clinical utility of sPIF in treating hypoxic-ischemic brain damage. The newly identified PIF/TLR4/KSRP/ let-7 regulatory axis also may operate during embryo implantation and development.

The effect of human cytomegalovirus (HCMV) on invasive capability of early pregnant extravillous cytotrophoblasts (EVTs) was investigated in vitro. Primary EVTs were obtained by complex phosphoesterasum digestion and gradient centrifugation from villous tissue aseptically taken from healthy pregnant women. Cytokeratin7 (CK7), vimentin (Vim) and c-erbB-2 were immunocytochemically detected to identify source of cells, and HCMVpp65 antigen was assayed to determine the infection state of primary EVTs by immunocytochemical staining. The EVTs were divided into two groups: control group and HCMV group, and the expression of c-erbB-2, matrix metalloproteinase-2 (MMP-2) and MMP-9 proteins was detected in two groups by immunocytochemistry and Western blotting. Enzymic activity changes of MMP-2 and MMP-9 were tested by gelatin zymography in primary EVTs infected with HCMV. The invasion of primary EVTs was detected by cell invasion assay in vitro after they were infected by HCMV. The cell source identification showed that the cells obtained were highly-pure primary EVTs, and primary EVTs could be infected by HCMV. Primary EVTs could express c-erbB-2, MMP-2 and MMP-9 proteins, and as compared with control group, the protein expression was decreased significantly in HCMV groups (P<0.05). Primary EVTs could secrete active MMP-2 and MMP-9 in vitro, and the activity of two MMPs was decreased significantly in HCMV groups (P<0.05). The in vitro cell invasion assay showed that the number of primary EVTs permeating Matrigel in HCMV group was decreased (P<0.05). We are led to conclude that HCMV can infect primary EVTs and inhibit their invasion capability, suggesting that the impaired EVT's invasion capability might be related to the abnormal expression of c-erbB-2, MMP-2 and MMP-9 proteins.

LeitthemaNeben der kontrovers diskutierten Präimplantationsdiagnostik mit Eingriff in die Integrität des Embryos sollen im Folgenden Möglichkeiten der nichtinvasiven Identifikation von Bio markern zur Beurteilung der embryonalen Entwicklungskompetenz diskutiert werden. Während die Analyse von Amnionflüssigkeit Aufschluss geben kann über mögliche Stoffwechselerkrankungen in einer bestehenden Schwangerschaft, ist es im Bereich der Reproduktionsmedizin denkbar, durch die Auswahl der besten Embryonen [in Deutschland durch das Embryonenschutzgesetz (ESchG) untersagt!] der möglichen Entstehung einer Erkrankung bereits vor dem Transfer vorzubeugen.Mit Vergabe des Nobelpreises an Professor Robert Edwards ist die künstliche Befruchtung erneut in das Interesse der breiten Öffentlichkeit getreten. Seit der Geburt von Louise Brown, dem ersten "Retortenbaby", im Jahr 1978 gibt es trotz rasanter Fortschritte in der Reproduktionsmedizin keine durchschlagenden Verbesserungen bei den Implantationsraten. Selbst bei der überwiegend im Ausland praktizierten Blastozystenkultur, die sich die natürliche Selektion der entwicklungskompetentesten Embryonen in vitro unter Zuhilfenahme morphologischer Kriterien zunutze macht, konnten keine eindeutigen Selektionsvorteile im Hinblick auf Schwangerschaftsraten nachgewiesen werden. Dies erklärt sich derzeit durch die geringeren Transferraten im Vergleich zu herkömm-lichen IVF/ICSI-Verfahren am Tag-2-oder -3-Transfer und durch schlechtere Schwangerschaftsraten nach Kryokonservierung von Blastozysten mit nachfolgendem Transfer [1], sodass andere Parameter herangezogen werden müssen. Proteomics and Secretomics in der ReproduktionsmedizinZiel neuerer Untersuchungen ist es deshalb, mittels Proteom-, Secretom-und Metabolomanalysen die frühestmögliche nichtinvasive Selektion des kompetentesten Embryos (. Tab. 1) zu erreichen, um idealerweise einen Single-EmbryoTransfer ohne Verlust an Schwangerschaftsraten bei gleichzeitiger Reduktion von Mehrlingsgraviditäten durchführen zu können. Einleitend muss jedoch festgehalten werden:Die Selektion entwicklungskompetenter Embryonen für den intrauterinen Transfer ist in Deutschland derzeit nicht mit dem aktuellen ESchG vereinbar. Die enge bidirektionale fetomaternale Kommunikation [2] wiederum unterstreicht die Relevanz der Sekretionsprodukte des Embryos im Hinblick auf die Implantationsfähigkeit. Die Tatsache, dass in der Reproduktionsmedizin Eizellen mit der umgebenden Follikelfüssigkeit gewonnen und Embryonen in vitro kultiviert werden, ermöglicht es, die Botenstoffe der Embryonen ohne weitere invasive Eingriffe zu analysieren. Denkbar wäre somit die Identifikation früher Sekretionsprodukte in der abpunktierten Follikel flüssigkeit. Doch um daraus auf einen einzelnen Embryonen schließen zu können, müssten sämtliche Follikelaspirate separat gewonnen werden, was mit erheblichem Mehraufwand bei der Punktion einhergeht.
