Prehypertensive Preconditioning Improves Adult Antihypertensive and Cardioprotective Treatment

Abstract: Transient prehypertensive treatment (TPT) causes prolonged antihypertensive and cardioprotective effects. Reduced angiotensin sensitivity has been attributed to those effects. We hypothesize that prehypertensive preconditioning by angiotensin II type 1 receptor (AT1R) blockade improves cardiovascular protection of late-onset AT1R blockade in a model of spontaneous hypertensive heart-failure rats (SHHF/Mcc-fa cp ). TPT (4 -8 weeks of age) consisted of AT1R blockade (5 mg/kg candesartan) or vehicle. Candesartan-… Show more

“…Agents that disrupt AT1R stimulation have profound protective effects in pathologically affected tissues relative to AngII signaling [60]. With the discovery of membrane associated estrogen receptors [61] and our findings here presented, 2ME2 has potential applications for the treatment of hypertension, or in fact any condition either caused or exacerbated by AngII stimulation of AT1R.…”

“…On the other hand, ATRAP-deficient mice have increased arterial BP (blood pressure) and an expanded PV (plasma volume) [49]. Essentially, ATRAP protects against cardiac hypertrophy and hypertension and it is worthy of note that transient prehypertensive treatment that prolongs anti-hypertensive and cardioprotective effects is likely to be beneficial owing to enhanced ATRAP expression [50].…”

“…Furthermore, an imbalance of AT1R and ATRAP in the myocardium is associated with the development of hypertension-associated LVH (14). However, there are few previously published studies on ATRAP expression in the myocardium of prehypertensive treated SHRs (15).…”
