Pregnancy-specific ?1-glycoprotein: cDNA cloning, tissue expression, and species specificity of one member of the PS?G family

Abstract: Pregnancy-specific beta 1-glycoprotein (PS beta G) is a heterogeneous product of the human syncytiotrophoblast, closely related to the CEA-NCA multigene family. In the present study, immunoscreening was carried out with anti-PS beta G antibodies to isolate cDNA sequences from a placental lambda gt11 expression library. One 1847-bp cDNA clone comprising the major portion of the coding sequence of a putative 48-kd peptide was sequenced and characterized. Hybridization of human genomic DNA to the PS beta G sequen… Show more

“…Many studies to determine expression of CEA-related genes at the transcriptional level have applied the Northern blot technique( 10, 44,54,57,58,61,65,[95][96][97][98][99][100][101][102][103][104][105][106][107]. Using this approach, and with the help of gene-specific or subgroup-specific DNA probes, a number of mRNA species varying in size have been identified in different tumors and some normal tissues.…”

“…We found that placental PS/3G represents a family of closely related glycoproteins of 72, 64, and 54 kDa and that placental poly(A)+ RNA directed the synthesis of three polypeptides of 50, 48 (major), and 36 kDa as immunoprecipitated by anti-PS3G serum (11). Moreover, near-full-length cDNAs encoding members ofthe PSf3G family were isolated and characterized by our laboratory (11,12) as well as by others (13)(14)(15)(16)(17)(18)(19)(20). Two gene fragments that share strong sequence similarity with the reported PSfG cDNAs have been reported (21,22).…”

“…Our PSBG! cDNA (Niemann et al, 1989) is nearly 100% homologous to PSBG sequences described previously (Streydio ct al., 1988; Watanabe and Chou. 1988) and contains a segment that is identical to the clone hPS3 published by …”
