Pregnancy resulting from intracytoplasmic injection of cryopreserved spermatozoa recovered from testicular biopsy

Abstract: We report a clinical pregnancy occurring in a 31 year old patient following intracytoplasmic sperm injection (ICSI) of cryopreserved spermatozoa obtained from a testicular biopsy. This was the couple's second attempt at an ovarian stimulated cycle resulting in the collection of 17 metaphase II ova which were all injected with progressively motile spermatozoa. A fertilization rate of 58% and a cleavage rate of 90% were achieved. This report is our first case of ICSI using cryopreserved testicular spermatozoa wh… Show more

“…Formal scrotal exploration was carried out before surgical extraction. The techniques followed those described in previous reports (17). The collected testicular samples also were stored in liquid nitrogen immediately for RNA extraction and sent for pathologic examination.…”

“…Cryopreservation of epididymal spermatozoa and testicular tissue before ICSI treatment is efficacious and may overcome many of the problems inherent to the use of fresh testicular tissue [16][17][18][19]. Additionally, the efficacy of cryopreserving testicular spermatozoa on the day of diagnostic testicular biopsy and the lack of significant differences in terms of fertilization and embryo developmental rates compared with fresh testicular spermatozoa have been reported [12].…”

“…Clinical pregnancies and delivery of healthy babies have been achieved with surgically retrieved epididymal [1][2][3][4] and testicular spermatozoa [5][6][7][8], but one of the major drawbacks lies in the need to 16 Urol Int 2000;65: [15][16][17][18][19][20] Trombetta/Liguori/Gianaroli/Magli/ Selman/Colpi/Belgrano/Vitali/Ferraretti resort to microsurgery at each ICSI attempt. Besides the patient's discomfort, repeated operations can damage epididymal and testicular tissues and, indeed, percutaneous epididymal sperm aspiration and testicular sperm aspiration were developed to recover sperm cells from the epididymis and testis without repeated surgical interventions [9,10].…”
