Predominant naturally processed peptides bound to HLA-DR1 are derived from MHC-related molecules and are heterogeneous in size

Abstract: Peptides bound to class I molecules are 8-10 amino acids long, and possess a binding motif representative of peptides that bind to a given class I allele. In the only published study of naturally processed peptides bound to class II molecules (mouse I-Ab and I-Eb), these peptides were longer (13-17 amino acids) and had heterogenous carboxy terminals but precise amino-terminal truncations. Here we report the characterization of acid-eluted peptides bound to HLA-DR1 by high-performance liquid chromatography, mas… Show more

“…The X-ray structure of a class II molecule [15] has confirmed that the groove is occupied by a single peptide, and also, that its structure is similar to the one found for class I molecules [16]. However, the binding site of class I molecules accommodates peptides of a length of 8 to 10 amino acids [17], whereas peptides eluted from class II molecules may vary between 8 to 23 amino acids [18].…”

“…The distribution of peptide size was normal (Figure 1), with sizes ranging from 9 to 43 amino acid length, most (88.7%) within the standard class II size range, 12 to 22 amino acids. 20,24 Peptides were derived from 192 different source proteins from several cellular compartments. To avoid redundancy, the most common protein localization was used for proteins belonging to more than one cell compartment.…”

“…Different studies have analyzed the peptide pools of several of these HLA-DR molecules (19,21,(35)(36)(37), but not in a comparative manner. The identification of structural features of the peptides bound to differentially RAassociated alleles is required to define the putative target of the T cell immune response generated by RA-related molecules.…”
