Predictive Value of Xrcc1 Gene Polymorphisms for Side Effects in Patients undergoing Whole Breast Radiotherapy: a Meta-analysis

Abstract: Our results suggest that XRCC1 399Gln and XRCC1 280Arg may be independent predictors of radiation-induced toxicity in post-surgical breast cancer patients, and the selection of genotyping method is an important factor in determining risk factors. No evidence for any predictive value of XRCC1 Arg194Trp and XRCC1 -77T>C was found. So, larger and well-designed studies might be required to further evaluate the predictive value of XRCC1 gene variation on radiation-induced side effects in patients undergoing whole b… Show more

“…Treatment-related factors include total RT dose, RT fraction size, RT technique, volume of skin irradiated, and addition of adjuvant chemotherapy. Patient-related factors include breast size, smoking history, diabetes mellitus, and BMI (Tucker et al, 1992;Fernando et al, 1996;Fisher et al, 2000;Pommier et al, 2004;Xie et al, 2012). In our study, total RT dose and lymph nodal irradiation were significant prognostic factors for radiation dermatitis.…”

“…In a recent meta-analysis [28], the predictive value of XRCC1 SNPs for side effects in patients undergoing whole breast radiotherapy has been determined. 11 studies mainly comprising Caucasian patients, addressing both acute and late toxicity, were included in the analysis.…”

“…Until now, no association between SNPs and late toxicity has been confirmed, either for BC or for prostate cancer [38] . Therefore, large, well-designed studies are needed to obtain more robust results.…”

“…The results of the meta-analysis by Xie et al [38] are summarised in Table 3. No significant association between the XRCC1 Arg399Gln SNP and IR-induced toxicity was observed in the overall analysis.…”
