Prediction of The Binding Mode of NP‐G2‐044 Targeting Fascin and Its Drug Resistance Mutations

Jinmei Pan,

Li‐Yan Xu
et al.

Abstract: Fascin is a major actin‐binding protein (ABP) in filopodia and is highly expressed in metastatic tumors. To inhibit its function, a potent inhibitor called NP‐G2‐044 is currently in phase II clinical trials. However, the binding mode of this inhibitor remains unclear. In this study, the binding mode of NP‐G2‐044 in fascin and predicted drug resistance mutations is investigated. The rough binding position and pose can be deduce based on information from its derivant NP‐G2‐029, whose crystal structure with fasci… Show more

“…Principal component analysis revealed an upward and rightward motion of WT-Fascin within domain 1, approaching domain 4 (supplementary material, Figure S8D). As NP-G2-044 binds to the cleft formed by domains 1 and 2 [27], this motion facilitated the creation of an accessible binding site for NP-G2-044 (supplementary material, Figure S8C). Conversely, Fascin K41R and Fascin K41Q mutants exhibited a distinct behavior, with a tendency for the cleft between domains 1 and 2 to close, rendering it unfavorable for NP-G2-044 binding to Fascin (supplementary material, Figure S8D).…”
