Prediction of Erosive Disease Development by Antimitochondrial Antibodies in Rheumatoid Arthritis

Abstract: Objective Mitochondria are found in the extracellular space in rheumatoid arthritis (RA). However, whether mitochondria are a source of autoantigens in RA has not been carefully addressed. Thus, we undertook this study to investigate the presence and significance of antimitochondrial antibodies (AMAs) in patients with RA. Methods AMAs were measured in serum samples from 3 cross‐sectional cohorts of RA patients (n = 95, n = 192, and n = 117) and healthy individuals (n = 38, n = 72, and n = 50) using a flow cyto… Show more

“…Moore et al hypothesised that anti-mitochondrial antibodies (AMAs) may be involved in the pathogenesis of RA [125] as extracellular mitochondria have been found in the plasma of RA patients [126]. Anti-mitochondrial antibodies were measured in three cohorts of RA patients and healthy controls, with disease progression being monitored over 8 years.…”

“…Furthermore, it was suggested that AMA levels could be used to predict erosive disease, independently of the "conventional" seropositive status (RF and ACPA). Additionally, antibodies that target the outer membrane protein of mitochondria (anti-mitofusin-1 antibodies) were found to be able to identify seronegative patients who were developing erosive disease [125].…”

“…Among them, specific antibodies against carbamylated hemopexin and alpha-2-macroglobulin allowed the diagnosis of 60% seronegative RA patients in the early arthritis cohort analysed. Another study investigating the presence and significance of antimitochondrial antibodies (AMA) found AMA levels to be elevated in up to 26% of RA patients (38). AMA levels correlated significantly with erosive disease and joint space narrowing, independent of ACPA positivity.…”
