Prediction of Anti-FVIII Inhibitor Persistence By Anti-FVIII IgG Subclasses in Patients with Severe Hemophilia — A in the Sippet Cohort Study

Abstract: Background: A major complication in severe hemophilia A is the formation of persistent neutralizing antibodies against factor(F) VIII, inhibitors, which render subsequent treatment ineffective. The presence of non-neutralizing anti-FVIII IgG antibodies (NNAs) before FVIII treatment initiation has been associated with subsequent development of inhibitors in previously untreated patients (PUPs) with severe hemophilia A in the frame of the SIPPET cohort (Cannavò A et al, Blood 2017). Up to 30% of P… Show more

“…Recently, a subanalysis of the SIPPET trial investigated the predictive value of IgG subclasses and the risk to develop a persistent or transient type of inhibitor (49). The concomitant presence of more than one IgG subclass and high-titer inhibitor was associated with a high risk to develop a persistent inhibitor.…”
