Pre- or post-treatment with aminoguanidine attenuates a renal distal acidification defect induced by acute ureteral obstruction in rats

Abstract: Acute unilateral ureteral obstruction (UUO) impairs distal nephron acid secretion and stimulates expression of inducible nitric oxide synthase (iNOS) in post-obstructed kidney (POK). This study investigated the influence of pre- or post-treatment with aminoguanidine as a selective iNOS inhibitor on UUO-induced renal functional disturbances. To induce acute UUO, the left ureter in rats was ligated and released after 24 h. Then, a 3 h clearance period followed by bicarbonate loading and thereafter a 30 min clear… Show more

“…Arterial oxygen tension (P a O 2 ), carbon dioxide tension (P a CO 2 ), acidity (pH a ), and bicarbonate concentration ([HCO 3 À ] a ) were measured by an Easy blood gas analyzer (Medica Corporation, Bedford, MA). 28,29 Plasma and urine samples were assayed for concentrations of creatinine, urea nitrogen, and glucose by means of an autoanalyzer (RA 1000; Technicon Instruments, Tarrytown, NY), concentrations of Na þ and K þ with an EasyLyte analyzer (Medica Corporation), and osmolality using a cryoscopic osmometer (Osmomat-010; Gonotec GmbH, Berlin, Germany). 29 Measurement of plasma malondialdehyde (MDA) and NO-metabolites…”

“…injected at 50 mg kg -1 at 6 h before and 9 h after unilateral renal ischemia or nephrectomy (Moosavi et al 2010;Moosavi et al 2011). Aminoguanidine hemisulfate (Molekula, Shaftesbury, Dorset, UK), a selective inhibitor of iNOS, was used at a dosage without affecting eNOS or nNOS (Moosavi et al 2013). It was intravenously infused at 20 mg kg -1 h -1 starting 21:30 h after unilateral renal ischemia or nephrectomy until the end of the experiment.…”
