Practical utility of insulin‐like growth factor II mRNA‐binding protein 3, glucose transporter 1, and epithelial membrane antigen for distinguishing malignant mesotheliomas from benign mesothelial proliferations

Abstract: The differentiation of malignant mesotheliomas and benign mesothelial proliferations is crucial in determining patient care and prognosis. But, this distinction can be extremely difficult, particularly in small biopsies. Recently, insulin-like growth factor II mRNA-binding protein 3 (IMP3) and glucose transporter 1 (GLUT-1) have been reported as specific and sensitive markers in the distinction of mesotheliomas from benign mesothelial proliferations. The purpose of this study is to evaluate the utility of IMP3… Show more

“…Of these, glucose transporter 1 (GLUT-1), a putative marker of malignancy, probably makes the most mechanistic sense, since it is increased in a very wide variety of malignant tumors. Unfortunately, GLUT-1 staining of mesothelial cells is confounded by staining of red cells, making interpretation difficult; and, more important, although some authors [5][6][7] claim that GLUT-1 staining is never seen in benign mesothelial reactions, our experience and that of others [8][9][10][11] ed positive for GLUT-1.…”

“…Immunohistochemistry was performed as reported previously 16,17 . Formalin-fixed, paraffinembedded tissues were cut into 4-µm-thick sections, deparaffinized in xylene, rehydrated by passage through decreasing concentrations of alcohol, and nally placed in distilled water.…”

“…GLUT-1 has been investigated by many groups for MPM differential diagnosis, and it has continuously shown high specificity for MPM (90-100%), while its sensitivity values ranged from 21% to 85% (5)(6)(7)(8).…”

“…The presence of an IMP-3 protein is indicative of MPM with a sensitivity of 37% to 94%, regardless of subtype and location. However, some benign reactions, particularly for atypical hyperplasia, stain for this marker as well (5,6,11,13).…”
