Zuojinwan (ZJW), a famous Chinese medicinal formula, contains two medicinal herbs Coptis chinese Frach and Evodia rutaecarpa (Juss.) Benth in the ratio of 6: 1. The inhibitory effects of ZJW on eight kinds of human cancer cell lines including SMMC-7721, BEL-7402, BEL-7404, HepG2, A549, NCI-H446, NCI-H460 and HCT-116 cells were evaluated, and the possible mechanism was investigated. The growths of the eight kinds of cancer cells were inhibited by ZJW assessed through MTT assay. Flow cytometry assay revealed a sub-G1 peak with reduced DNA content was formed. The cell cycle was arrested in the G0/G1 phase in ZJW-treated SMMC-7721 and HepG2 cells, and in the S phase for NCI-H460 cells. Significant DNA damage was produced by ZJW assessed with single-cell gel electrophoresis assay. Morphological changes were also observed. Caspase-3 and -9 activities were increased following ZJW treatment. Western blot analysis showed that Bax and Bak protein levels were increased after ZJW treatment, while Bcl-2 and Bcl-xl protein levels were decreased. Our results suggest that ZJW has significant anti-cancer activities due to induction of mitochondria-dependent apoptosis pathway. Therefore, ZJW has the potential to be a novel chemotherapy drug to treat hepatoma, lung cancer and colon cancer by suppressing tumor growth.

Our results demonstrated that S. umbellatum decreased the viability of EAT cells using both methods. Morphological analyses revealed that S. umbellatum-treatment induced EAT cell death by apoptotic pathway. We demonstrated the occurrence of reactive oxygen species (ROS) overgeneration, increased intracellular Ca(2+) concentration, alteration in mitochondrial membrane potential, phosphatydylserine externalization, and activation of caspases 3, 8, and 9. However, S. umbellatum produced myelotoxicity in bone marrow cells in a concentration-dependent manner. In comparison to EAT cells, the effects of S. umbellatum in bone marrow cells were 8-fold lower. Taken together, our results showed that S. umbellatum induced apoptosis in EAT cells at several levels and seems more toxic to tumor cells than to normal bone marrow cells.

RESUMO Este estudo foi conduzido para averiguar o uso de substratos orgânicos na produção de mudas nativas do Cerrado brasileiro. O objetivo foi avaliar os parâmetros biométricos, nutrição e qualidade de mudas de guapeva (Pouteria gardneriana) produzidas em substratos contendo diferentes fontes de matéria orgânica. Utilizaram-se os seguintes componentes para a formulação dos substratos: subsolo (SB), casca de arroz (RH), composto de esterco bovino [CC (silagem de milho + esterco bovino)], esterco bovino (CM), bagaço de cana (CB), torta de filtro de cana (FC) e BioPlant (BP). Cinco substratos foram formulados utilizando proporções em base de volume, como segue: SB+RH (1:1, v/v), SB+CC (1:1, v/v), SB+CM (3:1, v/v), CB+FC (3:2, v/v) e BP. Avaliou-se o comprimento do caule, o diâmetro do colo radicular, o peso seco da raiz, o peso seco da parte aérea, a nutrição e o índice de qualidade Dickson. A utilização do CM como fonte de matéria orgânica proporcionou, em geral, os melhores resultados para as características biométricas e a produção de matéria seca. O uso de matéria orgânica nos substratos SB+CM (3:1, v/v) e SB+CC (1:1, v/v) melhorou a qualidade do solo e favoreceu o crescimento e qualidade das mudas de guapeva, e podem ser indicados como substratos alternativos.
